摘  要:几丁质和壳聚糖是真菌细胞壁的重要组分,在维持细胞稳定性和寄主互作过程中发挥重要作用。为了分析不同发育阶段大丽轮枝菌(Verticillium dahliae Kleb.)细胞壁中几丁质和壳聚糖的构成,本研究以强致病力菌株V592为研究对象,依托CFW染色和壳聚糖抗体标记技术,对不同发育阶段大丽轮枝菌细胞壁中几丁质和壳聚糖的构成进行了显微观测。结果表明:大丽轮枝菌细胞壁中几丁质和壳聚糖的构成是一个动态变化的过程,除了分生孢子萌发点细胞壁以外,其它各个阶段的细胞壁中都有几丁质的存在,而壳聚糖主要存在于的分生孢子萌发点位置和芽管细胞壁上,另外侵染结构细胞壁上也存在壳聚糖。本研究对深入了解大丽轮枝菌细胞壁的发育生物学奠定了一定基础。Chitin and chitosan are important components of the fungal cell wall and play important roles in maintaining cell stability and host interaction.To analyze the composition of chitin and chitosan in the cell wall of Verticillium dahliae Kleb at different developmental stages,the strong pathogenic strain V592 was employed to observing the composition of chitin and chitosan in the cell wall of Verticillium dahliae at different developmental stages through microscope with CFW staining and chitosan antibody labeling technology.The results showed that the composition of chitin and chitosan in the cell wall of V.dahliae was varied in a dynamic process.Except for the cell wall of the conidial germination point,there is chitin in the cell wall at all other stages.Meanwhile,chitosan is mainly present at the location of the conidial germination point and the cell wall of the germ tube,and chitosan is also foundon the cell wall of the infected structure.This study provides a foundation for understanding the developmental biology of V.dahliae cell wall.
