摘 要:目的研究桑根酮C对3种肿瘤细胞增殖的抑制作用及其体内抗肿瘤作用。方法体外实验,选取3种癌细胞,桑根酮C体外共同培养,SRB法测桑根酮C对肿瘤细胞增殖的影响,计算IC 50。B16F0、B16F10细胞给予不同浓度桑根酮C,考察对B16F0、B16F10细胞内外黑素色含量的影响。体内实验,建立小鼠H22肿瘤动物模型,3个剂量(12.5、25、50 mg/kg)桑根酮C腹腔注射于小鼠体内,10 d后,取瘤组织和脏器,考察桑根酮C对荷瘤小鼠肿瘤生长和脏器指数的影响。结果桑根酮C抑制B16F0、B16F10、HepG2细胞增殖,其中HepG2最为敏感,IC 50为50.58±0.03μmol/L。不同浓度的桑根酮C都可使B16F0、B16F10细胞内外黑素色含量增加。桑根酮C高、中、低剂量对移植瘤小鼠肿瘤生长的抑制率为35%~47%,呈剂量依赖性;与模型组比较,均有显著性差异(P<0.05)。而且桑根酮C对荷瘤小鼠脏器指数影响非常小,与空白组比较没有显著性差异。结论桑根酮C对3种肿瘤细胞增殖有抑制作用,其在体内有抗肿瘤作用,且对小鼠脏器不良影响小。Objective To study the inhibitory effect of Sanggenon C on the proliferation of three tumor cells and its antitumor effect in vivo.Methods In vitro,three kinds of cancer cells were selected,Sanggenon C was cultured with cells.SRB method measured the effect of Sanggenon C on tumor cell proliferation and calculated IC 50.In vivo experiments,a mouse with H22 tumor cells was established,the animal models were treated with three concentrations of Sanggenon C(12.5,25,50 mg/kg)by ip.After administration for 10d,token the tumor tissue and viscera to investigate the effects of songgenon C on tumor growth and viscera index in tumor-bearing mice.Results Sanggenon C inhibited B16F0,B16F10,and HepG2 cell proliferation,of which HepG2 was the most sensitive,and IC 50 was 50.58±0.03μmol/L.The inhibition rates of tumor growth in transplanted mice by high,medium and low doses of Sanggenon C were 35%-47%,which were doses dependence.Compared with the model group,there were significant differences(P<0.05).In addition,the effect of Sanggenon C on the viscera index was very slight,and there was no significant difference with the blank group.Conclusion Sanggenon had inhibitory effect on the proliferation of three tumor cells and its antitumor effect in vivo.
