新疆石河子部分地区猪流行性腹泻病毒遗传变异分析

摘　　要：为分析猪流行性腹泻病毒(PEDV)遗传变异情况,从石河子的3个地区已确诊PED猪场采集病料,分别获得了3株PEDV S基因序列和ORF3基因序列,将其进行序列比对及遗传进化分析。根据3株S基因序列分析表明,XJ-SHZ148、XJSHZ146株与韩国QIAP1401株(KX793713)序列相似性最高,达到97.4%和98.7%;XJ-SW株与广东GD-03 2012株(KP870115)序列相似性最高,达到98.5%。3个分离株与PEDV疫苗株AF353511(CV777)性序列相似性分别为94.5%、94.2%和94.1%。系统进化分析表明,XJ-SHZ148、XJ-SHZ146与韩国的KPV1406(KM108353))、QIAP1401(KX793713))同为一群。XJ-SW株与广东GD/03/2012((KP870115))、GD/2011株(KP870115)同为一群;3株PEDV ORF3基因序列与疫苗株(CV777)序列比对多个位点存在突变,序列相似性较低。Porcine epidemic diarrhea causes an acute and highly contagious enteric disease in swine.To investigate the variation of PEDV,three sequences of S and ORF3 gene,respectively,were amplified from clinical samples in three pig farms that have been confirmed cases of PEDV.The S gene of XJ-SHZ148、XJ-SHZ146 is highest identity(97.4%,98.7%)with its homologue QIAP1401(KX793713)and the S gene of XJ-SW is highest identity(98.5%)with its homologue GD-03 2012(KP870115).The three sequence homology of proteins with the AF353511(CV777)were 94.5%、94.2%and 94.1%respectively.The phylogenetic tree shows that XJ-SHZ148、XJSHZ146 strain is closely related to KPV1406(KM108353)、QIAP1401(KX793713)strains from Korea,XJ-SW strain is closely related to GD/2011(KP870115)strains from China.The ORF3 consisted of 674 nucleotides,There were mutations at 7 positions between three isolates and the AF353511(CV777)strain,it showed a very low homology.
