摘  要:以玉米自交系昌72的花粉作为遗传转化受体,分别采用超声波处理法、电击法进行转化,比较2种方法处理后花粉的萌发率,以及电击转化后的结实率,探索玉米花粉介导的转基因方法在玉米转化中的应用。结果显示,超声波处理花粉的萌发率为19.01%,电击法处理花粉的萌发率为31.88%,电击法处理花粉的萌发率明显高于超声波处理方法。后期结实统计显示,电击处理后结实株数比例为75%,平均每株结实4颗。Maize pollens from maize self-pollinated line Chang 72 was used as the transformation target,and transformed by ultrasonic treatment and electroporation respectively.The germination rate efficiency of treated pollens were compared,and the application of maize pollen-mediated transgenic method was explored.Referring to previous method,the germination rate of pollen of ultrasonic treatment is 19.01%,the germination rate of electroporation pollen here is 31.88%,suggesting that the germination rate of electroporation pollen is significantly higher than that of ultrasonic treatment.The post-harvest statistics showed that the proportion of the number of sturdy plants after electroshock treatment was 75%,and the average number per plant is four.
