摘 要:为研究与泡桐丛枝病相关的长链非编码RNAs(lncRNAs),以毛泡桐(Paulownia tomentosa)的健康苗、患丛枝病的病苗及分别用100 mg·L^-1的利福平和60 mg·L^-1甲基磺酸甲酯处理病苗得到的处理苗为试验材料,利用高通量测序技术研究病健苗及试剂处理苗中的lncRNAs及相应靶基因的表达情况。共鉴定出3 700个lncRNAs,2 219个靶基因,从中得到389个差异表达的lncRNAs和对应的203个靶基因,并对其靶基因进行了GO功能分类。通过方案设计鉴定得到3个与泡桐丛枝病相关的lncRNAs,预测其对应的靶基因分别为磷脂酶D、蛋白磷酸酶以及WNK赖氨酸缺乏蛋白激酶。采用实时定量PCR技术验证随机挑选的5个lncRNAs及其靶基因,验证结果与高通量测序结果一致。In order to study long non-coding RNAs(lncRNAs)related to Paulownia witches’broom disease,using healthy seedlings,diseased seedlings and treated seedlings under the treatment of rifampicin 100 mg·L^-1 and methyl methanesulfonate 60 mg·L^-1 of Paulownia tomentosa as experimental materials,the expression of lncRNAs and its target genes in healthy seedlings and reagent-treated seedlings were studied by high-throughput sequencing technology A total of 3 700 lncRNAs and 2 219 target genes were identified,of which 389 differentially expressed lncRNAs and 203 target genes were identified.GO functional classification of target genes was carried out.Three lncRNAs related to Paulownia witches’broom were identified by scheme design,and the corresponding target genes were predicted to be phospholipase D,protein phosphatase and WNK lysine deficiency protein kinase.In addition,qRT-PCR technology was used to verify the random selection of five lncRNAs and their target genes.The results were consistent with high-throughput sequencing results.
