摘 要:采用PCR-DGGE技术对烟草土壤中细菌群落结构进行分析,对其中10条优势条带进行切胶回收、克隆测序及系统发育分析。DGGE图谱结果显示,施用多功能菌剂后细菌群落结构多样性比未施用多功能菌剂高。在采用多功能菌剂的前提下,减少化肥的施用量不会对烤烟的农艺性状和理化性质造成不良影响,T3处理(减肥10%)对土壤和烤烟的影响最明显。典范对应分析(canonical correspondence analysis,CCA)显示,pH、全氮和有效磷是造成烤烟土壤微生物群落结构变化的主要因素。PCR-DGGE(polymerase chain reaction-denaturing gradient gel electrophoresis)technology was employed to analyze the composition of bacterial community in selective different soil samples.The recovery,cloning,sequencing and phylogenetic analysis of 10 dominant bands were performed.The results of the DGGE showed that there was a difference in the diversity of bacteria between the soil samples with and without PGPR.Under the premise of use of microbial inoculants,reducing the use of fertilizers would not adverse effects on the agronomic traits and physicochemical properties of flue-cured tobacco.T3 treatment was most obvious impact on the soil and flue-cured tobacco.The canonical correspondence analysis(CCA)showed that soil pH,TN and A-P were the critical factors to shape the bacterial community structure in tobacco-growing soil.
