大鼠血清与心肌MMP-2、MMP-9、TIMP-1的相关性研究

摘　　要：目的研究大鼠血清与左室心肌组织中金属蛋白酶2、9(MMP-2、MMP-9)、金属蛋白酶组织抑制因子1(TIMP-1)水平的相关性。方法取材来源于本研究组前期实验中,将20只8周龄雄性原发性高血压大鼠(SHR)设为干预组,并随机分为2个亚组,每组10只,其中一组接受CD147(MMPs诱导剂)尾静脉注射,另一组除接受CD147注射外,加用多西环素(DOX,MMPs活性抑制剂)灌胃;另10只8周龄SHR及10只8周龄WKY大鼠设为非干预组,均予等剂量生理盐水尾静脉注射及灌胃。56 d后处死,取血清及左室心肌组织,通过ELISA及Western blot法分别检测血清与心肌组织MMP-2、MMP-9、TIMP-1水平,并统计血清与心肌组织MMP-2、MMP-9、TIMP-1的相关性。结果非干预组大鼠血清与左室心肌组织间TIMP-1及MMP-2/TIMP-1比值相关性较好,综合两组分析血清与左室心肌组织MMP-2/TIMP-1水平仍具明显相关性。结论大鼠血清与左室心肌组织中MMP-2/TIMP-1比值有较好的正相关趋势。Objective To study the correlation of metalloproteinase 2(MMP-2),MMP-9 and metalloproteinase tissue inhibitor 1(TIMP-1)between serum and left ventricular myocardial tissue in rats.Methods Eight weeks old male spontaneous hypertension rats(SHR)were selected as the intervention group from the preliminary experiment of our research group,which were randomly divided into 2 subgroups(n=10/subgroup).They were injected with CD147 by tail vein,or fed with doxycycline(DOX)by gavage simultaneously.The other eight weeks old SHR and WKY rats were chosen as the non-intervention group(n=10/group).They were all given equal dose normal saline by intravenous injection and gavage.The serum and left ventricular myocardial tissue were taken and the levels of MMP-2,MMP-9 and TIMP-1 were detected by ELISA and western blot when all the rats were killed 56 days later.The correlations of the above data were analyzed.Results The correlation between serum and left ventricular myocardial TIMP-1 and MMP-2/TIMP-1 ratio was good in the non-intervention group.Meanwhile,there was still a significant correlation between serum and left ventricular myocardial MMP-2/TIMP-1 in the comprehensive analysis in both groups.Conclusion The MMP-2/TIMP-1 ratio of serum has a significant positive correlation with left ventricular myocardial tissue in rats.
