摘 要:目的:研究复方红景天方对2型糖尿病肾病(Type 2 Diabetic Nephropathy,T2DN)的保护作用。方法:将雄性Wistar大鼠(SPF级,8周龄)50只随机分为正常对照组(N组)和模型组,模型组进行T2DN模型造模,造模成功的大鼠分为模型对照组(M组)、小剂量组(A组)、中剂量组(B组)、大剂量组(C组)。A、B、C组分别给予自拟中药方灌胃4周,治疗前后测量采取大鼠尾静脉血测空腹血糖(Glucose,GLU)、血胱抑素C(Cystatin C,CysC)及12 h尿微量白蛋白(Microalbumin,mALB)、体质量。结果:T2DN模型大鼠M组的血CysC、GLU、尿mALB明显高于N组(P<0.01)。M组的治疗前体质量高于N组(P<0.05),而M组的治疗后体质量明显低于N组(P<0.01)。M组治疗后体质量明显低于治疗前体质量(P<0.01)。治疗4周后M组体质量明显低于其他各组(P<0.01),模型组A、B、C组体质量无明显差异(P>0.05)。治疗4周后M组尿mALB、血CysC明显高于A、B、C组(P<0.01),M组GLU高于A、B、C组(P<0.05)。A、B、C组血CysC、尿mALB、GLU水平比较差异无统计学意义(P>0.05)。结论:复方红景天方可缓解T2DN模型大鼠体质量下降,降低T2DN大鼠血CysC、12 h尿mALB水平、血GLU水平,可以有效保护肾脏功能,其机制可以从延缓糖尿病肾病进展方面进一步研究。Objective:To observe the effects of the Fufang Hongjingtian prescription(复方红景天方)on kidney in rats with type 2 diabetic nephropathy.Methods:Fifty male Wistar rats with 8–weeks-old were randomly divided into the normal control group(N group)and model group.During the 45 rats,43 rat models with DN were successfully made.Then,they were randomly divided into the model control group(M group),low-dose group(A group),medium dose group(B group)and high dose group(C group).Rats in the A、B and C group were treated by 3、6 and 12 g/(kd-1·d-1)prescription and rats in the model control group were treated by intragastric administration of the same amount of normal saline for 4 weeks.Results:Blood CysC,GLU and urine mALB in group M were significantly higher than those in N group(P<0.01).Pre-treatment weight in group M was higher than N group(P<0.05),while posttreatment weight in group M was significantly lower than N group(P<0.01).The weight in group M after treatment was significantly lower than before treatment(P<0.01).After 4 weeks,the weight in group M was significantly lower than other groups(P<0.01),and there was no significant difference in the weight among model group A,B and C(P>0.05).After 4 weeks,urine mALB and blood CysC in group M were significantly higher than those in group A,B and C(P<0.01),and GLU in group M was significantly higher than those in group A,B and C(P<0.05).There was no statistically significant difference in blood CysC,urine mALB and FPG levels among groups A,B and C(P>0.05).Conclusion:The Fufang Hongjingtian prescription can alleviate the weight loss of T2DN model rats,and reduce the blood CysC,12 h urine mALB level and blood GLU level,which can effectively protect renal function,and its mechanism can be further studied from the perspective of delaying the progression of diabetic nephropathy.
