摘  要:主要探讨了三种生长因子对猪卵母细胞成熟及孤雌胚胎发育及多能基因表达的影响,结果表明:成熟液中添加生长因子后可显著提高卵母细胞成熟率;成熟液和发育液中都添加生长因子后胚胎卵裂率、囊胚率及胚胎质量均显著升高;添加生长因子可极显著提高孤雌囊胚中多能基因的表达.本研究为提高猪卵母细胞成熟及胚胎早期发育奠定了基础,进而为获得更利于分离培养猪胚胎干细胞系的囊胚材料提供了参考.The effects of three growth factors on the maturation of porcine oocytes,the development and pluripotent genes expression of porcine early parthenogenetic embryos were analyzed in this study.The results showed that when the growth factors were supplemented in the oocyte in vitro maturation media,the maturation rate could be significantly increased.When the growth factors were supplemented in both in vitro maturation media and porcine zygotes medium 3(PZM-3),the cleavage rate and blastocyst rate,as well as the quality of the parthenogenetic embryos were significantly increased.Moreover,the growth factors could significantly improve the expression level of pluripotent genes OCT4 and SOX2 in parthenogenetic blastocysts.This study provided a useful reference in improving the maturation of porcine oocytes and the early embryonic development,and will be also helpful for isolation and establishment of porcine embryonic stem cell lines.
