摘 要:目的评估金银花水煎剂体外逆转产金属酶铜绿假单胞菌耐药性能力强弱,为中药抗感染治疗提供有益借鉴。方法质粒接合转移试验验证耐药性R质粒存在,倍比稀释法确定试验菌株对金银花水煎剂的最小抑菌浓度,以次最小抑菌浓度作用于试验菌株进行R质粒消除试验,平板影印法确认消除子并计算消除率。结果23株产金属酶铜绿假单胞菌临床分离株,有7株接合转移试验阳性,占30.43%(7/23);均为blaIMP型,未发现blaVIM型;7株试验菌株对金银花水煎剂的MIC为321.4 mg/ml;金银花水煎剂和5%SDS对7株试验菌株在24、48和72 h的R质粒平均消除率分别为0、0.71%、0.62%和0.24%、1.33%、1.09%。结论金银花水煎剂可在体外通过R质粒消除的方式逆转产金属酶铜绿假单胞菌耐药性,逆转效率处在低水平,不及SDS。OBJECTIVE To evaluate the reversal ability of the Lonicera japonica decoction on the resistance of metallo-β-lactmases producing Pseudomonas aeruginosa in vitro,so as to provide helpful reference for the anti-infection treatment using TCM.METHODS Conjugation test was used to examine the presence of R plasmid,and broth double dilution method was used to determine the minimum inhibitory concentration of L japonica decoction on the tested strains.Sub minimum inhibitory concentration was used for R plasmid elimination experiment in the tested strains,and replica plating was conducted to verify the elimination of the plasmid and calculate the eliminating rate.RESULTS Among the 23 clinical siolates of metallo-β-lactmases producing P aeruginosa,7 strains were positive in conjugation experiment,accounting for 30.43%(7/23).The genotype was all bla IMP,and no bla VIM was detected.The average minimum inhibitory concentration of the 7 strains to L japonica decoction was 321.4 mg/mL.The average elimination rate of R plasmid in the 7 strains by L japonica decoction and 5%SDS at 24 h,48 and 72 h were 0,0.71%,0.62%and 0.24%,1.33%,1.09%,respectively.CONCLUSION Lonicera japonica decoction can reverse the resistance of metallo-β-lactmases producing P aeruginosa by R plasmid elimination in vitro,and the reversal efficiency is lower than SDS.
