摘  要:目的研究一贯煎对肝纤维化大鼠的治疗机制。方法大鼠分为正常对照组、模型组、阳性对照组和中药组。通过肝功能检测、HE染色、Masson染色评测各组大鼠的肝纤维化程度。通过免疫组织化学法检测各组大鼠肝组织中Notch1蛋白在细胞质中表达量的变化。结果与正常对照组相比,模型组大鼠肝功能指标谷丙转氨酶(alanine transaminase,ALT)、谷草转氨酶(aspartate transaminusei,AST)明显增高(P<0.05),Notch1表达增加(P<0.05);与模型组相比,阳性对照组和中药组大鼠肝功能指标ALT、AST明显降低(P<0.05),Notch1表达降低(P<0.05),肝组织纤维化明显好转。结论一贯煎能明显改善肝纤维化,可通过抑制Notch1的表达达到治疗肝纤维化的效果。Objective To study the therapeutic mechanism of Yiguan Decoction on liver fibrosis in rats.Methods Rats were divided into normal control group,model group,positive control group and traditional Chinese medicine group.Liver fibrosis degree of each group was evaluated by liver function test,HE staining and Masson staining.Expression of Notch1 protein in cytoplasm of rats’liver tissue in each group was detected by immunohistochemistry.Results Compared with the normal control group,the liver function indexes alanine transaminase(ALT)and aspartate transaminase(AST)of the model group were significantly increased(P<0.05),and the expression of Notch1 was increased(P<0.05).Compared with the model group,the liver function indexes ALT and AST of the positive control group and the traditional Chinese medicine group were significantly decreased(P<0.05),the Notch1 expression was decreased(P<0.05),and the liver fibrosis was significantly improved.Conclusion Yiguan Decoction can significantly improve liver fibrosis,and it achieves the effect of treating liver fibrosis by inhibiting the expression of Notch1.
