滋水清肝理冲饮对子宫内膜异位症小鼠应用促性腺激素释放激素激动剂后性激素表达的影响

摘　　要：目的观察滋水清肝理冲饮对子宫内膜异位症小鼠应用促性腺激素释放激素激动剂(gonadotropin releasing hormone agonists,GnRHa)后血清雌激素(estradiol,E 2)、促黄体生成素(luteinizing hormone,LH)、卵泡刺激素(follicle stimulating hormone,FSH)水平的影响,并通过对其在位、异位内膜组织中免疫因子白介素6(interleukin-6,IL-6)、与血管内皮生长因子(ascular endothelial growth factor,VEGF)的检测验证其应用的安全性。方法雌性BALB/c小鼠,随机分为正常组、疾病模型组、GnRHa组、中药高剂量组、中药中剂量组、中药低剂量组,采用ELISA法检测给药后各组小鼠血清E 2、LH、FSH,采用免疫组化法检测各组免疫因子的表达。结果与疾病模型组相比较,GnRHa组小鼠血清FSH、LH含量升高,E 2含量下降(P<0.01);与GnRHa组相比较,中药高、中剂量组FSH、LH含量下降,E 2含量升高(P<0.01)。与疾病模型组相比,应用GnRHa后在位、异位内膜组织IL-6、VEGF表达降低;中药各组干预后未出现IL-6、VEGF表达升高的情况。结论从各组小鼠血清E 2、LH、FSH浓度水平结果可见,GnRHa皮下注射后,小鼠体内FSH、LH含量升高,E 2含量下降,而高、中剂量滋水清肝理冲饮可改善子宫内膜异位症小鼠应用GnRHa引起的体内性激素改变。并且,改善GnRHa导致副作用的同时,不会引起子宫内膜异位症免疫功能紊乱,安全性良好。Objective To observe the effects of Zishui Qinggan Lichong Decoction on serum levels of estradiol(E 2),luteinizing hormone(LH)and follicle stimulating hormone(FSH)in endometriosis(EMT)mice after application of gonadotropin releasing hormone agonists(GnRHa)and to verify the safety of its application by detecting immune factors(interleukin-6,IL-6)and vascular endothelial growth factors(VEGF)in eutopic and ectopic endometrium.Methods BALB/c female mice were randomly divided into normal group,disease model group,GnRHa group,high dose of Chinese medicine group,medium dose of Chinese medicine group and low dose of Chinese medicine group.After giving the medicine,ELISA method was used for detecting serum levels of E 2,LH and FSH in the groups of mice,and the expression of immune factors in each group was detected by immunohistochemistry.Results By contrast with the disease model group,the serum levels of FSH and LH in the GnRHa group were significantly increased,while that of E 2 was significantly decreased(P<0.01).The serum levels of FSH and LH in the high dose of Chinese medicine group and the medium dose of Chinese medicine group were significantly decreased,while that of E 2 was significantly increased(P<0.01)in contrast with the GnRHa group.Compared with disease model group,the expressions of IL-6 and VEGF in eutopic and ectopic endometrial tissues were decreased after GnRHa treatment;there was no increase of IL-6 and VEGF expressions in each Chinese medicine group after the intervention.Conclusion From the results of serum levels of E 2,LH and FSH in groups of mice,after subcutaneous injection of GnRHa,the serum levels of FSH and LH in mice were increased,and the serum level of E 2 was decreased.However,high and medium doses of Zishui Qinggan Lichong Decoction can improve the change of sex hormone in EMs mice undergoing GnRHa.At the sametime,Zishui Qinggan Lichong Decoction can improve side effects caused by GnRHa,and does not cause immune dysfunction in endometriosis,with good safety.
