摘 要:目的观察龙贝逍遥散冻干粉对人乳腺癌细胞株MDA-MB-231增殖、迁移的作用及对凋亡通路相关蛋白的影响。方法CCK8法检测人乳腺癌细胞MDA-MB-231的增殖抑制率,划痕实验检测人乳腺癌细胞MDA-MB-231迁移,q PCR测定细胞P53、BCL-2的表达。结果与对照组相比,龙贝逍遥散冻干粉处理48 h和72 h时能够浓度依赖性的抑制人乳腺癌细胞MDA-MB-231的生长,表现为OD值逐步降低(P<0.05),龙贝逍遥散冻干粉半数抑制乳腺癌细胞增殖的浓度(IC50),48 h为0.761 3 mg/ml,72 h为0.766 4 mg/ml;相同条件培养48 h后,0 mg/ml(对照组)、0.7 mg/ml、0.8 mg/ml的龙贝逍遥散冻干粉培养后MDA-MB-231细胞覆盖区域占起始划痕面积的百分比分别为76.38%、64.87%、66.52%,加入龙贝逍遥散冻干粉后,MDA-MB-231的迁移能力明显减弱(P<0.05),差异均有统计学意义。与未经龙贝逍遥散冻干粉处理的MDA-MB-231细胞相比,经龙贝逍遥散冻干粉处理的MDA-MB-231细胞P53表达较高,BCL-2表达较低(P<0.05)。结论龙贝逍遥散能抑制乳腺癌细胞的增殖与迁移,其机制与调节P53/BCL-2凋亡信号通路有关。Objective To observe the effects of freeze-dried powder of longbei xiaoyao san on MDA-MB-231 proliferation and migration of human breast cancer cell line and the expression of P53 and BCL-2.Methods CCK8 method was adopted to determine the inhibition rate of MDA-MB-231 proliferation in human breast cancer.The scratch test was used to determine the migration of MDA-MB-231 and qPCR was to determine the expressions of P53 and BCL-2.Results Compared with the control group,In 48h and 72 h after the treatment with the freeze dried powder of longbei xiaoyao san,MDA-MB-231 growth was inhibited in concentration-dependence and OD value was reduced gradually(P<0.05).The 50%inhibitory concentration of freeze-dried powder of longbei xiaoyao san(IC50)was 0.7613 mg/ml in 48 h and was 0.7664 mg/ml in 72 h.In 48h under the same culture condition,the percentages of MDA-MB-231 coverage zone in the initial scratch area were 76.38%,64.87%and 66.52%in 0 mg/ml group(control group),0.7 mg/ml group and 0.8 mg/ml group of freeze-dried powder of longbei xiaoyao san successively.After the freeze-dried powder of longbei xiaoyao san added,the migration of MDA-MB-231 was weakened obviously(P<0.05),indicating the statistical significance.Compared with MDA-MB-231 without treated with the freeze-dried powder of longbei xiaoyao san,P52 expression was higher and BCL-2 was lower for the cell treated with the freeze-dried powder(P<0.05).Conclusion Longbei xiaoyao san inhibits the proliferation and migration of breast cancer cell,which is related with the regulation of P53/BCL-2 apoptotic signaling pathway.
