摘 要:目的研究通络化痰胶囊对MCAO模型大鼠脑缺血损伤后皮层区域神经轴索抑制因子勿动蛋白(Nogo-A)及轴突再生促进因子生长相关蛋白43(GAP-43)表达的影响。方法48只Wistar大鼠随机分为正常组、模型组、通络化痰胶囊高、低剂量组。除正常组外,其余各组参照Koizumi法制作大鼠大脑中动脉缺血模型(MCAO)。高、低剂量组造模清醒后分别给予648 mg/kg、324 mg/kg通络化痰胶囊2 ml。各组大鼠同期观察7 d、14 d。应用免疫组化法检测Nogo-A、GAP-43表达水平。结果与模型组相比,通络化痰胶囊高、低剂量组均能降低Nogo-A表达(P<0.05,P<0.01),上调GAP-43的表达(P<0.05)。结论通络化痰胶囊可通过降低Nogo-A的表达、上调GAP-43的表达,改善脑缺血区域神经轴突再生的微环境,从而发挥其脑保护作用。Objective To study the effect of tongluo huatan capsules on the expressions of Nogo-A and GAP-43 in the cortex after ischemia injury in MCAO model rats.Methods A total of 48 Wistar rats were randomized into a normal group,a model group,a high-dose group and a low-dose group of tongluo huatan capsules.In reference to Koizumi method,MCAO model was prepared in the rats.The IHC method was adopted to determine the expressions of Nogo-A and GAP-43.Results Compared with the model group,in the high-dose and the low-dose group of tongluo huatan capsules,Nogo-A expression was reduced(P<0.05,P<0.01)and GAP-43 was down-regulated(P<0.05).Conclusion Tongluo huatan capsules improve the microenvironment of axon regeneration in cerebral ischemic area,thus play the role to the brain protection.
