油用牡丹根结线虫拮抗菌株的筛选和鉴定

摘  要:油用牡丹是一类重要的木本粮油树种。随着种植面积的扩大,根结线虫病发生日益严重。为了安全有效的防治油用牡丹根结线虫病,本文从土壤中分离获得了1株对油用牡丹根结线虫具有拮抗效果的菌株HZ087,经形态特征观察、生理生化特性测定以及16S rDNA序列分析,鉴定该菌株为解淀粉芽孢杆菌Bacillus amyloliquefaciens。利用该菌株发酵液5倍、10倍稀释液处理油用牡丹根结线虫2龄幼虫,48h后校正死亡率分别为91.36%、83.53%;利用发酵液2倍、5倍稀释液处理油用牡丹根结线虫卵,10d后孵化率为45.98%、49.27%,可以有效抑制油用牡丹根结线虫卵的孵化。Oil tree peony is an economically important woody oil tree in China.With the expansion of cultivated area of oil tree peony,the root-knot nematode is also more and more prominent,and the peony seeds production is limited.In order to control the root-knot nematode with safe and environmentally friendly,the bacterium isolate HZ087 was obtained from the soil,and it showed the highest nematicidal activity to Meloidogyne hapla.Isolate HZ087 was identified to be Bacillus amyloliquefaciens with morphological observation,physiological and biochemical test and 16S rDNA sequence analysis.Second stage Meloidogyne hapla were treated with 5-fold and 10-fold diluents of fermentation liquor of isolate HZ087 for 48h in vitro methods,the corrected mortality were 91.36%、83.53%respectively.The eggs were treated with 2-fold and 5-fold diluents of fermentation liquor for 10 days,the egg hatchability were 45.98%and 49.27%.
