摘  要:为探究中华绒螯蟹(Eriocheie sinensis)肝胰腺坏死综合征(HPNS)的病因,利用PCR-DGGE技术分析了健康蟹、肝胰腺坏死综合征病蟹、传染性皮下及造血组织坏死病毒(IHHNV)感染蟹、白斑综合征病毒(WSSV)感染蟹及IHHNV-WSSV共感染蟹的肝胰腺菌群结构。结果显示,肝胰腺坏死综合征病蟹组与IHHNV-WSSV共感染蟹组首先聚类(相似度0.72),随后依次与IHHNV感染组(相似度0.69)及WSSV感染组(相似度0.62)聚类,最后与健康蟹组聚类(相似度0.56),表明肝胰腺坏死综合征病蟹的肝胰腺菌群结构与IHHNV-WSSV共感染蟹的较为一致,与健康蟹的差异明显。此外,肝胰腺坏死综合征病蟹、WSSV感染蟹、IHHNV感染蟹、IHHNV-WSSV感染蟹与健康蟹相比,肝胰腺菌群多样性降低,肝胰腺菌群中气单胞菌属、假单胞菌属等致病菌的丰度增加,鞘氨醇单胞菌属、鞘氨醇杆菌属等有益菌的丰度减少,甚至肝胰腺坏死综合征病蟹中未出现慢生根瘤菌属。初步推论,HPNS的发生可能与IHHNV-WSSV共感染有关,IHHNV-WSSV共感染改变了河蟹肝胰腺菌群结构,可能引发肝胰腺正常的生理功能下降,继而停止了摄食行为,从而逐渐发展形成HPNS初期的肝胰腺白化现象。PCR-DGGE technology were used to analyze the hepatopancreas microflora structure of crabs in healthy,crabs with HPNS,crabs infected with infectious hypodermal haematopoietic necrosis virus(IHHNV),crabs infected with white spot syndrome virus(WSSV)and crabs co-infected by IHHNV-WSSV in order to explore the etiology of Hepatopancreatic necrosis syndrome(HPNS)in Eriocheie sinensis.The result showed that the HPNS group clustered with the group of co-infection by IHHNV-WSSV firstly(similarity of 0.72),then clustered with the group of infection by IHHNV(similarity of 0.69)and WSSV(similarity of 0.62)in sequence,and finally clustered with healthy crab(similarity of 0.56).It suggested that the hepatopancreas microflora structure of HPNS crab was similar with that of crab co-infected by IHHNV-WSSV,while it was obviously different with that of healthy crab.In addition,the Shannon-Wiener index of hepatopancreas microflora of HPNS disease crabs,crabs infected with WSSV,crabs infected with IHHNV,crabs co-infected with by IHHNV-WSSV were lower than that of healthy crabs.The abundance of pathogenic bacteria such as Aeromonas,Pseudomonas in hepatopancreatic microflora increased.The abundance of available bacteria such as Sphingomonas and Sphingobacterium decreased,even Bradyrhizobium was not shown in HPNS disease crabs.It was preliminarily deduced that the occurrence of HPNS might be related to the IHHNV-WSSV co-infection.The IHHNV-WSSV co-infection changed the hepatopancreas microflora structure,which might cause the decline of physiological function of hepatopancreas,made crabs stop feeding,and then gradually developing the albinism of hepatopancreas in the early stage of HPNS.
