复方蜥蜴散不同微粒组合剂含药大鼠血清对人SGC-7901胃癌细胞SIRT1、P53的影响

摘　　要：目的:研究复方蜥蜴散不同微粒组合剂含药SD大鼠血清对人SGC-7901胃癌细胞SITR1、P53表达水平的影响及机制。方法:160只SPF级雄性SD大鼠随机分8组,每组20只。各组分别予相应药物干预1周后,于腹主动脉处取血,离心,制备含药血清,加于人SGC-7901胃癌细胞培养24h,Western-Blot检测人SGC-7901胃癌细胞SIRT1、P53表达水平。结果:(1)80目+100目高剂量组表达低于其他药物组(P<0.05)。(2)P53蛋白表达比较:复方蜥蜴散各组表达均低于华蟾素片阳性对照组、5-Fu阳性对照组(P<0.05);80目+100目混合组表达低于80目组、100目组(P<0.05);80目+100目高剂量组的表达低于80目+100目中、低剂量组(P<0.05)。结论:复方蜥蜴散可通过干预SIRT1、P53蛋白表达,抑制细胞增殖,促进细胞凋亡。Objectiv:To investigate the effect of SD rat's serum containing different microparticle combined agents of compound lizard powder on the expressions of SITR1 and P53 in human gastric cancer cells SGC-7901,and to explore mechanism.Methods:160 male SD rats of SPF were randomLy divided into 8 groups:normal saline negative control group,compound lizard powder 80 mesh group,100 mesh group,80 mesh+100 mesh high-,medium-and low-dose group,cinobufacin tablet positive control group and 5-Fu positive control group,20 rats in each group.After each group was given corresponding drug intervention for 1 week,blood was taken from the abdominal aorta,centrifuged and prepared into drug-containing serum.The drug-containing serum was cultured in human gastric cancer cells SGC-7901 for 24 hours.The expression levels of SIRT1 and P53 in human gastric cancer cells SGC-7901 were detected by Western-Blot.Results:1.Comparison of SIRT1 protein expression:the expression of 80 mesh+100 mesh high-dose group was lower than that of other drug groups,(P﹤0.05).2.Comparison of P53 protein expression:the expression of each compound lizard powder group was lower than that of cinobufacin tablet positive control group and 5-Fu positive control group(P﹤0.05);the expression of 80 mesh+100 mesh mixed group was lower than that of 80 mesh group and 100 mesh group(P﹤0.05);the expression of 80 mesh+100 mesh high-dose group was lower than that of 80 mesh+100 mesh medium-and low-dose group(P﹤0.05).Conclusion:Compound lizard powder can inhibit cell proliferation and promote cell apoptosis by interfering with expressions of SIRT1 and P53 protein.
