摘 要:目的研究不同强度的工频电磁场对体外培养的小鼠海马神经元HT22细胞增殖、凋亡及磷酸化c-Jun氨基末端激酶(p-JNK)和磷酸化细胞外信号调节激酶(p-ERK)表达的影响,探索工频电磁场的生物安全性。方法将正常培养的HT22细胞随机分成4组,其中1组为对照组,不作任何处理,其余3组为实验组,分别曝露于强度为10 mT、20 mT和30 mT的工频电磁场,20 min/d,连续3 d。MTS法检测各组细胞增殖情况,Annexin V-FITC/PI及流式细胞术检测各组细胞凋亡情况,Western blotting检测各组p-JNK和p-ERK的表达情况。结果与对照组相比,实验组均促进细胞增殖,其中20 mT组促进增殖作用最强(P<0.01);与对照组相比,实验组未见明显凋亡情况;实验组p-JNK和p-ERK表达均高于对照组,其中20 mT组p-JNK和p-ERK表达显著增高(P<0.05)。结论10 mT、20 mT和30 mT工频电磁场曝露可以促进HT22细胞增殖,增殖率与电磁场强度呈非线性关系,其机制可能是通过JNK和ERK的磷酸化水平进行调节。Objective To study the effects of power-frequency electromagnetic fields on the proliferation,apoptosis,and expression of phosphorylated c-Jun N-terminal kinase(p-JNK)and phosphorylated extracellular signal regulated kinase(p-ERK)in cultured HT22 cells in vitro and to explore the biological safety of power-frequency electromagnetic fields.Methods Normal cultured HT22 cells were randomly divided into four groups:one control group without treatment and three experimental groups exposed to power-frequency electro-magnetic fields of 10 mT,20 mT,and 30 mT,respectively,for 20 minutes every day for three days.MTS was used to detect cell prolifera-tion.Annexin V-FITC/PI and flow cytometry were used to detect cell apoptosis.Western blotting was used to detect the expression of p-JNK and p-ERK.Results HT22 proliferation was higher in the experimental groups than in the control group.Compared with the control group,the experimental groups had no obvious apoptosis.The expression of p-JNK and p-ERK in the experimental groups was higher than that in the control group and the expression of p-JNK and p-ERK in 20 mT group increased significantly(P<0.05).Conclusion Pow-er-frequency electromagnetic fields may promote HT22 cell proliferation through phosphorylation of JNK and ERK,and the proliferation rate has a non-linear relationship with magnetic field intensity.
