摘  要:目的探讨神经化学物质对大鼠负性注意偏向的影响。方法应用不可预知慢性温和应激(CUMS)建立大鼠抑郁症模型(实验组)。对实验组和对照组大鼠进行注意偏向测试;采集2组大鼠海马附近神经组织,利用高效液相色谱检测神经递质5-羟色胺(5-HT)、5-羟(基)吲哚乙酸(5-HIAAA)、去甲肾上腺素(NE)、3,4-二羟苯乙酸(DOPAC)、内啡肽(EP)、高香草酸(HVA)水平;分析神经递质水平与注意偏向的相关性。结果CUMS抑郁症大鼠模型造模成功;实验组大鼠出现了抑郁症状;实验组大鼠出现了明显的负性偏向;实验组大鼠5-HIAA、DOPAC水平明显高于对照组,5-HT、EP和HVA水平则明显低于对照组,而NE水平无统计学差异;实验组大鼠5-HIAA和HVA与注意偏向的变化有显著关联,5-HT和DOPAC水平的变化对注意偏向有影响。结论CUMS抑郁症模型适用于大鼠。抑郁症大鼠神经递质水平明显不同于正常大鼠,神经递质对抑郁症大鼠的注意偏向有直接影响。Objective To investigate the causal effect of neurotransmitters on negative attentional biases.Methods In a randomized controlled trial,chronic unpredictable mild stress(CUMS)was used to create a depressed rat model,as the experimental group.After the attentional biases tests were conducted on the experimental and control groups,the nervous tissues around the hippocampi of all the rats were excised to detect neurotransmitters,such as 5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAAA),norepinephrine(NE),3,4-dihydroxyphenylacetic acid(DOPAC),endorphin(EP),and homovanillic acid(HVA).The correlations between the scores of the attentional biases and neurotransmitters were analyzed.Results The CUMS rat model was successfully created.All the rats in the experimental group exhibited depressed behavior.The concentration of 5-HIAA and DOPAC in the experimental group were higher than those of the control group,while the concentration of 5-HT,EP,and HVA in the experimental group were lower than those of con-trol group,and the level of NE was the same in all rats.The correlations between 5-HIAA,HVA,and attentional biases were significant,suggesting that neurotransmitters(i.e.5-HT and DOPAC)significantly contribute to the level of negative attentional biases.Conclu-sion CUMS can successfully induce a depressed rat model.The level of neurotransmitters in the depressed rats is different from those of normal rats.Moreover,neurotransmitters have a relationship with attentional biases of rats.
