摘  要:目的:建立家兔腹膜组织中5-Fu含量的测定方法。方法:采用C 18柱(Φ4.6×200 mm,5μm),甲醇-乙酸-水(3∶0.05∶96.95)为流动相,流速1.0 mL/min,波长254 nm,柱温30℃进行检测。结果:该方法测得腹膜中5-Fu的浓度在0.011~0.660 mg/g组织(n=9;r=0.999 9)范围内与其峰面积有良好的线性关系;高、中、低3个浓度的回收率分别为102.00%、98.07%、115.10%,RSD为1.08%、0.31%、0.57%。结论:该方法操作简便,灵敏度高,结果准确可靠,适用于5-Fu在家兔腹膜组织中药物浓度的测定。Objective:To develop a method for the determination of 5-fluorouracil concentration in peritoneal tissue of rabbits.Methods:The C 18 column(Φ4.6×200 mm,5μm)was used with methanol-acetic acid(99%)-water(3∶0.05∶96.95)as the mobile phase.The flow rate was 1.0 mL/min,the detective wavelength was 254 nm,the column temperature was 30℃.Result:A good linear relationship was obtained between the peak area and the concentration of fluorouracil from 0.011 to 0.660 mg/g tissue(n=9;r=0.999 9).The recoveries were 102.00%,98.07%,115.10%and the relative standard deviations was 1.08%,0.31%,0.57%in the high,middle,and low concentrations,respectively.Conclusion:A simple,sensitive and selective method for the determination of fluorouracil in peritoneal tissue of rabbits was established.
