摘 要:目的:获取白花丹参Cu/Zn SOD基因的全长cDNA序列,并分析其序列特征和不同外源胁迫下的表达特征。方法:利用已有白花丹参cDNA全长文库,随机测序获得SmCu/Zn SOD全长序列。BLAST进行序列比对,Motif Scan软件分析氨基酸序列的功能区段,RT-PCR技术分析基因表达特征。结果:获得ORF为459 bp的SmCu/Zn SOD全长cDNA序列,编码氨基酸152个,氨基酸序列分析发现其具有典型的Cu/Zn SOD PROSITE pattern,RT-PCR分析其在叶片中表达量最高。水分亏缺、盐胁迫、低温和ABA处理均能较强的诱导该基因的表达。结论:本研究首次获得白花丹参Cu/Zn SOD全长基因序列,该基因可能参与对干旱、盐碱和低温等非生物胁迫的抵抗。Objective:To obtain the full-length cDNA sequence of Cu/Zn SOD gene of Salvia miltiorrhiza f.alba,and to analyze its sequence characteristics and expression characteristics under different exogenous stresses.Methods:SmCu/Zn SOD full-length sequences were obtained by random sequencing using the existing full-length cDNA library of Salvia miltiorrhiza f.alba.BLAST was used for comparation,Motif Scan software was used to analysis the functional segment of amino acid sequence.RT-PCR was used to detect the gene expression characteristics.Results:SmCu/Zn SOD full-length cDNA sequence with ORF of 459 bp was obtained and putatively encoded a polypeptide of 152 amino acids.Amino acid sequence analysis revealed that it had a typical Cu/Zn SOD PROSITE pattern,RT-PCR analysis showed the highest expression in leaves.Water deficit,salt stress,low temperature and ABA treatment could strongly induce the expression of this gene.Conclusion:The Cu/Zn SOD full-length gene sequence of Salvia miltiorrhiza f.alba is obtained for the first time,the gene may be involved in the resistance to abiotic stresses such as drought,salinity and low temperature.
