摘　　要：目的探讨老龄急性心肌梗死(acute myocardial infarction,AMI)小鼠心肌组织转录因子叉头框蛋白O(forkhead box protein O,FoxO)1、FoxO3a蛋白及氧化应激指标表达变化。方法3月龄小鼠12只,随机分为低龄观察组、低龄对照组各6只;18月龄小鼠12只,随机分为老龄观察组、老龄对照组各6只。低龄观察组、老龄观察组结扎左前降支制备AMI模型,低龄对照组、老龄对照组仅开胸不结扎冠状动脉。造模成功后24h,应用血流动力学检测仪检测4组小鼠血流动力学指标;处死小鼠取心肌组织,采用Western blot法检测FoxO1和FoxO3a蛋白相对表达量,ELISA法检测活性氧(reactive oxygen species,ROS)、一氧化氮(nitric oxide,NO)、丙二醛(malondialdehyde,MDA)、总抗氧化能力(total antioxidant capacity,T-AOC)水平及超氧化物歧化酶(superoxide dismutase,SOD)活性。结果造模成功后24h,低龄对照组、老龄对照组、低龄观察组、老龄观察组左心室舒张压[(70.7±3.1)、(62.3±2.1)、(58.3±3.5)、(49.3±1.5)mm Hg]、左心室内压最大上升速率[(3 050.3±90.3)、(2 584.0±68.4)、(2 142.3±99.0)、(1 813.7±57.1)mm Hg/s]、左心室内压最大下降速率[(3 846.7±124.6)、(3 434.7±66.7)、(2 045.7±79.8)、(1 798.3±69.7)mm Hg/s]、FoxO1相对表达量(0.62±0.02、0.39±0.03、0.32±0.02、0.23±0.01)、FoxO3a蛋白相对表达量(0.62±0.01、0.43±0.03、0.36±0.02、0.23±0.02)及SOD活性[(363.91±11.13)、(266.40±11.33)、(219.05±6.80)、(103.52±10.18)u/mL]、T-AOC[(4.59±0.38)、(3.92±0.31)、(2.79±0.16)、(2.05±0.17)u/mg]依次降低(P<0.05),NO[(12.08±0.51)、(14.56±0.45)、(24.49±0.32)、(31.62±0.40)μmol/L]、ROS[(35.69±1.06)、(44.64±1.54)、(105.56±1.09)、(131.75±2.62)u/mL]及MDA水平[(161.32±4.17)、(288.52±15.71)、(412.86±5.10)、(709.00±10.73)μmol/L]依次升高(P<0.05);低龄观察组、老龄观察组心率[(402.7±14.6)、(361.3±9.5)次/min]较低龄对照组[(482.0±23.6)次/min]、老龄对照组[(444.3±11.0)次/min]慢(PObjective To investigate the expressions of forkhead box protein O(FoxO)1,FoxO3aand oxidative stress related indicators in the myocardium of aged mice after acute myocardial infarction(AMI).Methods Twelve 3-month old mice were divided into young observation group(n=6)and young control group(n=6),and another 12aged mice were divided into aged observation group(n=6)and aged control group(n=6).The left anterior descending artery was ligated to establish AMI models in young and aged observation groups,while young and aged control groups only received open chest surgery without ligating the coronary artery.The hemodynamic indexes were detected in 4groups 24hafter modeling.The myocardium was obtained after the mice were sacrificed to detect the relative expressions of FoxO1and FoxO3aby Western blot,while the activity of reactive oxidative species(ROS),the levels of nitric oxide(NO),malondialdehyde(MDA)and the total antioxidant capacity(T-AOC),as well as the activity of superoxide dismutase(SOD)were detected by ELISA.Results In young control group,aged control group,young observation group and aged observation group 24hafter modeling,the left ventricular diastolic blood pressure((70.7±3.1),(62.3±2.1),(58.3±3.5),(49.3±1.5)mm Hg),the maximal rate of the increase of left ventricular pressure((3 050.3±90.3),(2 584.0±68.4),(2 142.3±99.0),(1 813.7±57.1)mm Hg/s)and the maximal rate of decrease of left ventricular pressure(3 846.7±124.6),(3 434.7±66.7),(2 045.7±79.8),(1 798.3±69.7)mm Hg/s),as well as the relative expression levels of FoxO1(0.62±0.02,0.39±0.03,0.32±0.02,0.23±0.01),FoxO3a(0.62±0.01,0.43±0.03,0.36±0.02,0.23±0.02),SOD activity((363.91±11.13),(266.40±11.33),(219.05±6.80),(103.52±10.18)u/mL)and T-AOC((4.59±0.38),(3.92±0.31),(2.79±0.16),(2.05±0.17)u/mg)decreased gradually in turn(P<0.05);NO((12.08±0.51),(14.56±0.45),(24.49±0.32),(31.62±0.40)μmol/L),ROS((35.69±1.06),(44.64±1.54),(105.56±1.09),(131.75±2.62)u/mL)and MDA((161.32±4.17),(288.52±15.71),(412.86±5.10),(709.00±10.73)μmo
