摘 要:目的探讨4种不同破红液在血性胸水富集瘤细胞行苏木精伊红(HE)染色及免疫细胞化学(ICC)染色中的效果。方法选取肺腺癌血性胸水样本22例,肉眼观察外观呈深红色或褐色,离心无法分层。分别用不同的破红液(1%乙酸液,50%乙醇液,50%乙醇与10%中性缓冲甲醛混合液,1%乙酸与50%乙醇及10%中性缓冲甲醛混合液)进行破红,观察HE染色及ICC染色结果,并进行比较及评估。结果4组破红液均能消除红细胞,使肿瘤细胞显现,但肿瘤细胞在形态学和抗原表达方面,50%乙醇与10%中性缓冲甲醛混合液富集瘤细胞的染色效果明显优于另外3种破红液(P<0.001)。结论不同的破红液对血性胸水富集瘤细胞的HE染色及ICC染色效果不尽相同,采用50%乙醇与10%中性缓冲甲醛混合液进行破红的效果最佳。Objective To explore the effects of Hematoxylin&Esoin(HE)staining and immunocytochemistry staining(ICC)of the enrichment tumor cell of hemorrhagic pleural effusion treated with 4 different kinds of red blood cell(RBC)lysis buffer.Methods We chose 22 cases of crimson or brown hemorrhagic pleural effusion caused by adenocarcinoma of lung,and these sample couldn′t be stratified when centrifuged.We used different RBC lysis buffers including 1%acetic acid,50%ethanol,the mixture of 50%ethanol and 10%neutral buffer formalin(NBF),the mixture of 1%acetic acid,50%ethanol and 10%NBF to lyse the RBC,and we assessed the results of HE and ICC.Results All of 4 kinds of RBC lysis buffer could eliminate the RBCs and made the tumor cells observable.The mixture of 50%ethanol and 10%NBF was better than the other three in the aspect of cellular morphology and antigen expression(P<0.001).Conclusion Enrichment tumor cell treated with different RBC lysis buffers has different effects of HE and ICC.The mixture of 50%ethanol and 10% NBF has the best effect,and may have application value,but it still needs more samples and a multicenter study.
