摘 要:为鉴定北京地区枸杞(Lycium chinense Mill)白粉病的病原,采用形态学对其病原菌有性世代闭囊壳形态进行显微观察,同时利用分子生物学方法克隆其rDNA ITS序列,在此基础上构建进化树,并分析其亲缘关系。结果表明,该白粉菌闭囊壳直径110.5~168.4μm,附属丝较多并生于闭囊壳的“赤道”部位,长度为60.4~220.4μm,其顶端1~3次二叉或三叉状分支;BLASTn显示其ITS序列(MG757562.1)与GenBank中的穆氏节丝壳(Arthrocladiella mougeotii(Lév.)Vassilk)的ITS序列同源性大于99.8%,而与其他白粉菌ITS序列的同源性小于91.0%;进化分析显示其与穆氏节丝壳中国菌株(JX546296.1、KP420475.1)、日本菌株(AB022380.1)、韩国菌株(AF455748.1)、土耳其菌株(KX017568.1)、澳大利亚菌株(AF073358.1)聚为一支,亲缘关系最近。因此,引起北京地区枸杞白粉病的病原菌鉴定为节丝壳属(Arthrocladiella)的穆氏节丝壳(A.mougeotii)。Disease investigation revealed that powdery mildew was serious in Lycium chinense Mill in Beijing.In order to identify the pathogen,the morphology of cleistothecium of powdery mildew sexual generation is observed and the ITS(ribosomal DNA internal transcribed spacer)regions are amplified and sequenced.Based on the ITS sequences,the phylogenetic tree is constructed and the genetic relationship of selected powdery mildews is analyzed.The results show as follow:the diameter of the cleistothecium of the powdery mildew is 110.5~168.4μm,and its appendages are abundant and equatorial with 60.4~220.4μm length and 1~3 bifurcate or trifurcate branches;the ITS sequence(MG757562.1)shared more than 99.8%homology with that of Arthrocladiella mougeotii in GenBank,but less than 91.0%homology with those of other powdery mildews;in the phylogenetic tree,the powdery mildew is clustered in the same clade with some strains of A.mougeotii isolated from L.chinense,such as KP420475.1(China),AB022380.1(Japan),AF455748.1(Korea),KX017568.1(Turkey),JX546296.1(China)and AF073358.1(Australia).Based on these results,the pathogen of the powdery mildew infecting L.chinense in Beijing is identified as A.mougeotii.
