童子一号草莓组织培养快繁体系的建立

摘　　要：为提高草莓种苗的繁殖速度及质量,以童子一号的茎尖为外植体,研究适宜的生长、丛生芽增殖的培养基、继代时间、继代次数以及生根培养基,并探讨了适宜的移栽条件。结果表明:草莓茎尖生长培养基为1.0 mg·L^-1 6-BA+0.2~0.5 mg·L^-1 NAA;增殖培养基为MS+2 mg·L^-1 6-BA+0.1 mg·L^-1 NAA,每30~40 d继代一次,可连续继代4次;生根培养基为0.2~0.5 mg·L^-1 IBA+0.2 mg·L^-1 NAA;组培苗根系长至2 cm,植株高4 cm时进行移栽,移栽后遮光率不高于60%、空气湿度不高于60%、环境温度高于20℃,移栽成活率可达到87%。In order to improve the speed and quality of strawberry breeding,in this experiment,the stem tip of Tongzi 1 strawberry was used as explants,we studied suitable for growth of medium,effects of plant growth regulator on multiple shoot clumps proliferation,effects of transgenerational days and number on adventitious bud proliferation,the best culture medium and transplanting conditions for tissue culture seedlings.The results showed that the strawberry stem tip germination medium for 1.0 mg·L^-1 6-BA+0.2-0.5 mg·L^-1 NAA;Best adventitious bud proliferation culture medium for MS+2 mg·L^-1 6-BA+0.1 mg·L^-1 NAA,once 30-40 d,it could be successively relayed for 4 times;The best rooting culture medium for MS+0.2-0.5 mg·L^-1 IBA+0.2 mg·L^-1 NAA;We transplanted tissue culture seedling,when it grew to 2 cm of root long and 4 cm of plant height;After transplanting shading coefficient was not higher than 60%,humidity of the air was not higher than 60%,environment temperature was above 20℃,transplanting survival rate could reach 85%.
