摘  要:近年来,柯萨奇病毒A组10型(Coxsackievirus A10,CV-A10)逐渐成为手足口病(Hand,foot and mouth disease,HFMD)最重要的病原体之一。相比肠道病毒A组71型(Enterovirus 71,EV-A71),CV-A10尚未有上市的疫苗,其构象表位也缺乏系统性研究。本研究利用实验室发展的人肠道病毒构象表位的生物信息学预测算法,对CV-A10的构象表位进行系统性预测。结果显示:45个氨基酸残基聚集成三簇构象表位:site 1、site 2和site 3。site 1位于病毒衣壳表面峡谷的北侧,site 2和site 3位于峡谷南侧,三者呈现"品"字形分布。site 2和site 3与已报道的两个表位2G8和VP2-P28高度重叠,说明预测结果具有较高准确性。鼻病毒B种14型(Rhinovirus B14,RV-B14)、脊髓灰质炎病毒1型(Poliovirus type 1,PV1)和CV-A10同属于人肠道病毒。已有研究表明,RVB14和PV1的抗原表位也呈三簇分布,且每一簇都与CV-A10的预测结果高度重叠。这既说明了预测结果的准确性,同时也提示三簇构象表位的分布模式可能是人肠道病毒构象表位的基本分布规律。CV-A10构象表位的预测结果,为构象表位的实验鉴定、病毒的分子流行病学研究和疫苗研发提供了重要支持。Coxsackievirus A10(CV-A10)is one of the main causative agents of hand,foot and mouth disease(HFMD). Compared with enterovirus A71(EV-A71),there is no approved vaccine or systematic research for the conformational epitopes of CV-A10. A bioinformatics-based algorithm for human enterovirus,developed by our research team,was used to predict the conformational epitopes of CV-A10. We found that 45 amino-acid residues were clustered into three sites:site 1,site 2 and site 3. Site 1 was located on the north rim of the canyon,whereas site 2 and site 3 were located on the south rim of the canyon on the surface of CV-A10.Epitopes were distributed in three sites as the Chinese character " 品 ". Site 2 and site 3 overlapped highly with two epitopes 2 G8 and VP2-P28,indicating that the prediction results were accurate. Rhinovirus B14(RVB14),poliovirus type 1(PV1)and CV-A10 belonged to human enterovirus. The epitopes of RV-B14 and PV1 were also clustered into three sites,and each site overlapped highly with the predicted results of CV-A10,which not only proved the accuracy of the prediction results,but also suggested that the three sites distribution pattern of conformational epitopes among human enteroviruses was present. Our prediction results of CV-A10 can provide essential support for experimental identification of conformational epitopes,molecular epidemiologic studies and vaccine development.
