摘 要:AP2/EREBP是植物特有的一类具有庞大家族的转录因子。该转录因子广泛参与调节植物生理功能和水杨酸、茉莉酸、脱落酸和乙烯等信号转导途径。本研究利用艾纳香的全长转录组数据,通过生物信息学的手段对艾纳香AP2/EREBP家族进行挖掘并分析,为从分子水平研究艾纳香代谢调控提供参考依据。利用DNAMAN、MEGA5.05软件以及Protparam、SOPMA在线网站,对艾纳香AP2/EREBP转录因子家族的类型、结构域、系统进化、蛋白理化性质和氨基酸序列高级结构等进行分析。结果表明从艾纳香转录组中挖掘到57个AP2/EREBP基因,通过查找开放阅读框筛选出18条能翻译成完整蛋白的序列;根据结构域的划分可分为DREB、ERF和AP2三个亚族;通过与拟南芥AP2/EREBP相关基因进行系统进化分析,结果表明艾纳香DREB和ERF亚族成员分别属于B2、B4、B5、B6和A6簇;生物信息学表明艾纳香AP2/EREBP转录因子家族蛋白富含酸性氨基酸,热稳定性较高且均为亲水性蛋白,全部蛋白都以无规则卷曲为主;每个亚家族的蛋白质三级结构差异不明显,但不同亚族之间有差异,主要表现在β-转角、β-折叠及无规则卷曲长度不同。本研究结果为进一步研究艾纳香AP2/EREBP基因功能提供参考。AP2/ERE B P is a plant-specific transcription factor with a large family.This transcription factor is widely involved in regulating plant physiological functions and signal transduction pathways such as salicylic acid,jasmonic acid,abscisic acid and ethylene.In this study,the full-length transcriptome database of Blumea blasamifera was used to excavate and analyze the members of AP2/EREBP family by bioinformatics,which provided a reference for studying the metabolic regulation of Blumea blasamifera at the molecular level.DNAMAN,MEGA5.05 software and Protparam,SOPMA online website were used to analyze and predict the types,domains,phylogenetic,physicochemical properties,high-level structures of amino acid sequence of the AP2/EREBP transcription factor in Blumea blasamifera.The results showed that there were 57 AP2/EREBP genes in Blumea blasamifera genome,and 18 consecutive translated protein sequences were screened by looking up the open reading frame.According to the division of the domain,they could be divided into DREB,ERF and AP2 three subfamilies.The phylogenetic analysis of AP2/EREBP related genes in Arabidopsis thaliana showed that members of DREB and ERF subfamilies in Blumea blasamifera belonged to B2,B4,B5,B6 and A6 clusters respectively.And the bioinformatics indicates that the AP2/EREBP family proteins of Blumea blasamifera were hydrophilic proteins,with high thermal stability and rich in acid amino acids.Meanwhile,all proteins were dominated by random coils,the tertiary structure of proteins in each subfamily was not obvious,but there were differences among different subfamilies,mainly inβ-turn,extended strand and random coil.The results of this study provide a reference for further research on the function of AP2/EREBP gene in einechion.
