摘 要:本研究利用甲基磺酸乙酯(ethyl methane sulfonate,EMS)诱导黄瓜"YZ-01"发生突变,以构建黄瓜突变体库,从而获得纯合突变体,加速功能基因的解析进程。通过对比5组不同浓度EMS(0,0.5%,1%,1.5%,2%和2.5%)处理后黄瓜种子的出苗率,最终确定该品系的半致死剂量为1.5%。在此基础上进行了突变体库的构建,EMS诱变处理3 100粒种子,最终成功定植1 340株植株;其中93个植株收到M1代自交种子;将M1每个株系选择8到10株种植,其中88个M2株系成活;M2植株表型突变频率为15.22%。对M2各株系表型鉴定,获得了一系列重要突变性状,例如矮化突变体、果皮黄化突变体、果皮光亮突变体、短果突变体、圆叶突变体、叶缘失绿突变体、黄叶突变体、叶片皱缩突变体等。该研究为今后黄瓜功能基因研究及遗传育种改良提供理论基础。In this study,ethyl methane sulfonate(EMS)was used to induce mutation of cucumber"YZ-01"to construct a cucumber mutant library,thereby obtaining homozygous mutants and accelerating the process of functional gene analysis.By comparing the emergence rate of cucumber seeds treated with 5 different concentrations of EMS(0,0.5%,1%,1.5%,2%and 2.5%),the semi-lethal dose of the line was determined to be1.5%.On this basis,the construction of the mutant library was carried out.3 100 seeds were treated by EMS mutagenesis,and 1 340 plants were successfully planted;93 plants received self-crossing seeds of M1 generation;8 to 10 plants were selected to planted from M1 generation,88 M2 strains survived;phenotypic identification of M2 lines,obtained a series of important mutation traits,such as dwarf mutant,peel yellowing mutant,peel bright mutant,short fruit mutant,round Leaf mutants,leaf margin chlorotic mutants,yellow leaf mutants,leaf shrub mutants,and petal and calyx conjoined mutants.The phenotypic mutation frequency of M2 plants was 15.22%.This study provided the base theory for future research on cucumber functional genes and genetic breeding.
