摘 要:为了明确甲基磺酸乙酯(ethy methan sulfonate,EMS)溶液浓度及处理时间对白桦种子发芽时间、发芽率以及幼苗生长的影响,本研究以白桦种子为供试材料,设置4个处理时间(3 h,5 h,7 h,10 h)及6个EMS浓度梯度(0%,0.2%,0.4%,0.6%,0.8%,1.0%)对白桦种子进行诱变处理。结果表明:0.2%~1.0%的EMS溶液浸泡处理3~10 h会延迟白桦种子发芽,浓度越高所需初始发芽时间越长;1.0%EMS诱变处理的种子初始发芽平均需要6.92 d,是对照组5.25 d的1.32倍,7 h处理的种子所需的起始发芽时间最长,平均为6.45 d;EMS诱变处理显著降低了白桦种子发芽率,浓度越高处理时间越长发芽率越低;EMS溶液浓度由0%增加到1.0%时,平均发芽率由9.72%降低到3.61%,其中处理时间为10 h时发芽率仅为3.33%;EMS诱变处理显著抑制了幼苗生长,导致茎长和根长明显变短。根据种子发芽及幼苗生长情况,本研究确定EMS诱变处理白桦种子的适宜条件为1.0%处理5 h或0.4%处理7 h。In order to clarify the effect of ethy methan sulfonate(EMS)solution concentration and treatment time on the germination time,germination rate and seedling growth,the seeds of Betula platyphylla were treated with the combination of four treatment time(3 h,5 h,7 h,10 h)and six EMS concentration gradients(0%,0.2%,0.4%,0.6%,0.8%,1.0%).The results showed that the germination time of seeds were delayed after soaked in 0.2~1.0%EMS solution for 3~10 h.As the concentration was higher,the initial germination time was much longer.When the concentration of EMS was up to 1.0%,the initial germination needed 6.92 d,which was 1.32 times as long as control group 5.25 d.The seeds treated with EMS solution for 7 h showed the longest initial germination time,which was 6.45 d on average.EMS mutagenic treatment significantly reduced the germination rate of seeds.The higher the concentration and longer the treatment time,the lower was the germination rate.When the concentration of EMS solution increased from 0 to 1.0%,the average germination rate decreased from 9.72%to3.61%.The germination rate was only 3.33%,when the treatment time was 10 h.EMS mutagenic treatment significantly inhibited seedling growth.Stem length and root length of seedlings significantly shortened.According to the seed germination and seedling growth,the suitable conditions of EMS mutagenesis were treatments with1.0%concentration for 5 h or 0.4%for 7 h.
