摘 要:为了探索百合不同杂种系之间的遗传多样性和亲缘关系,本研究对7个百合杂种系96个品种开展了ISSR分子标记研究,从30个引物中筛选出25个引物,将重复性良好的引物用于百合不同杂种系品种的ISSR分析,其中共扩增出121条带,其中多态性条带共107条,多态性比率达88.43%。通过Popgene 1.32分析百合不同杂种系之间的遗传距离,其中最近的是A和LA (0.031 0);最远的是LO和OT (0.269 2),7个百合杂种系之间的遗传相似系数在0.969 4~0.764 0之间,Nei’s基因多样性指数(He)和Shannon信息指数(I)分别是0.333 0和0.502 4,说明百合品种存在丰富的遗传变异。采用UPGMA聚类分析将7个百合杂种系分为两大类,OA系列品种单独为一类,剩余6个杂种系被分为一类。此外,ISSR条带分析显示,不同引物扩增下,L、O、A杂种系内所有品种存在一些特征条带,并且可作为该杂种系的指示条带。这些条带在LA、LO、OT和OA杂种系中品种也得到初步验证。本研究对百合不同杂种系品种进行ISSR标记分析遗传多样性和亲缘关系,探索百合品种的遗传多样性,以期为百合杂种系之间新品种杂交育种时的亲本选配提供依据。In order to explore the genetic diversity and genetic relationship among different hybrid lines of lily,ISSR molecular markers were studied in 96 varieties of 7 lily hybrids in this study. 25 primers were screened from30 primers, and reproducible primers were used for ISSR analysis of different hybrids of lily. There were 121 bands, of which 107 were polymorphic bands, and the polymorphism ratio was 88.43%. The genetic distance between different hybrid lines of lily was analyzed by Popgene 1.32, the most recent of which was A and LA(0.031 0);the farthest was LO and OT(0.269 2), and the genetic similarity coefficient between 7 lily hybrids was0.969 4~0.764 0, Nei’s gene diversity index(He) and Shannon information index(I) were 0.333 0 and 0.502 4 respectively, indicating that there are abundant genetic variations in lily varieties. Seven lily hybrid lines were divided into two groups by UPGMA cluster analysis. The OA series were individually divided into one group, and the remaining six hybrid lines were divided into one group. In addition, ISSR band analysis showed that there were some characteristic bands in all varieties of L, O, A hybrids under different primer amplification, and could serve as indicator bands for the hybrid lines. These bands were also initially validated in the LA, LO, OT, and OA hybrid lines. In this study, ISSR markers were used to analyze the genetic diversity and genetic relationship of different hybrid cultivars of lily, and the genetic diversity of lily cultivars was explored, in order to provide a basis for the selection of parents in hybrid breeding of hybrids between lily hybrids.
