摘 要:目的:建立侧柏炭TLC鉴别、指纹图谱和含量测定方法,提升侧柏炭质控标准。方法:采用二次展开法对侧柏炭中异槲皮苷、槲皮苷、穗花双黄酮、槲皮素和山奈酚进行TLC鉴别;采用HPLC法建立侧柏炭指纹图谱;采用HPLC法同时对侧柏炭中槲皮苷、槲皮素、山奈酚、穗花双黄酮4种黄酮类成分进行含量测定。结果:10批侧柏炭中在与异槲皮苷、槲皮苷、穗花双黄酮、槲皮素和山奈酚相应的位置上均显相同颜色的荧光斑点;10批侧柏炭指纹图谱中标识了14个共有峰,并指认2、3、7、8、11号色谱峰分别对应为异槲皮苷、槲皮苷、槲皮素、山奈酚和穗花双黄酮。建立的HPLC法可同时对侧柏炭中槲皮苷、槲皮素、山奈酚、穗花双黄酮进行含量测定,简便可行。结论:本研究可用于全面提升侧柏炭饮片质量标准,以进一步有效保障其临床疗效。Objective: The TLC identification, fingerprint and content determination were established to improve the quality control standard of Platycladi Cacumen carbonisata pieces. Methods: Using double-developing method of TLC to identify isoquercitroside, quercitroside, amentoflavone, quercetin and kaempferol in Platycladi Cacumen carbonisata pieces;fingerprint was established by using HPLC method;The contents of 4 flavonoids(quercitroside, quercetin, kaempferol and amentoflavone) were determined by HPLC method. Results: Fluorescent spots of 10 batches of Platycladi Cacumen carbonisata pieces showed the same color fluorescent spots on the corresponding position of isoquercitroside, quercitroside, amentoflavone, quercetin and kaempferol;14 common peaks were identified in the fingerprint, and 2, 3, 7, 8, 11 peaks were respectively isoquercitroside, quercitroside, quercetin, kaempferol and amentoflavone. A simple and feasible method for the simultaneous determination of quercitroside, quercetin, kaempferol and amentoflavone was developed. Conclusion: This study can be used to improve the quality standard of Platycladi Cacumen carbonisata pieces, and which can furtherly guarantee its clinical efficacy.
