摘 要:目的:探讨鹿茸蛋白对缺血缺氧条件下心肌细胞活力的影响。方法:鹿茸蛋白处理方法同前报道。由SD大鼠乳鼠心脏组织中收集、培养心肌细胞,分为对照组、模型组和不同浓度鹿茸蛋白干预组。通过对细胞形态的观察及心肌细胞活性的检测,筛选缺血缺氧的条件及鹿茸蛋白浓度。进一步应用EdU试剂盒检测心肌细胞DNA活性。结果:36h缺血缺氧能够较好的制备心肌细胞损伤模型,鹿茸蛋白的配制浓度确定为0.25、0.5、1mg/mL。缺血缺氧干预后,鹿茸蛋白0.25、0.5、1mg/mL组细胞的活力明显优于模型组(P<0.01);与对照组比较,模型组心肌细胞DNA活性显著下降(P<0.01),鹿茸蛋白1mg/mL组心肌细胞的DNA活性显著升高(P<0.01)。结论:鹿茸蛋白能够减轻36h缺血缺氧造成的心肌细胞损伤。Objective: To investigate effect of velvet antler protein on the myocardial cell activity suffering from ischemiahypoxia. Methods: The approach of velvet antler proteins was reported earlier. Enzymolytic method and differential attachment were suitable for culture of myocardial cell from cardiac tissue of SD lactating rats. The experiment was divided into control group, model group and intervention group with different concentrations of pilose antler protein. Myocardial cell activity was detected by MTS kit and EdU kit, and the duration of ischemic-hypoxia and velvet antler proteins concentration were screened. Results: 36 h ischemia and hypoxia could make a better model of myocardial cell injury. The configuration concentration of velvet antler protein was determined to be 0.25, 0.5, 1 mg/mL. Compared with model group, Velvet antler proteins( 0.25, 0.5, 1 mg/mL) treatment resulted in an improvement in cell activity(P<0.01);and also 1 mg/mL velvet antler proteins could maintain cardiomyocytes DNA replication(P<0.01), and 1 mg/mL pilose antler protein could significantly increase the DNA activity of cardiac myocytes(P<0.01). Conclusion: Velvet antler proteins regulate myocardial cell injury from 36 hours ischemic-hypoxia.
