补肾益气活血方对去势大鼠Wnt7b/β-catenin信号通路的调控机制  被引量:19

摘  要:目的:基于Wnt7b/β-catenin信号通路研究补肾益气活血方对绝经后骨质疏松症大鼠股骨的作用及影响。方法:将90只雌性SD大鼠随机分成正常组,模空组,补肾益气活血方高、低剂量组和福善美组,每组18只。除正常组外,其余各组大鼠采用一次性摘除大鼠双侧卵巢法建立骨质疏松症动物模型,正常组、模空组大鼠给予0.9%氯化钠溶液灌胃,其余各组给予相应药物灌胃,共灌胃12周后,取材。采用双能X线吸收测量法(DAX)检测股骨骨密度;采用ELISA法检测血清ALP、TRACP水平以及股骨Wnt7b、Axin2、Runx2蛋白表达水平;采用qRTPCR法检测股骨Wnt7b、Axin2、Runx2 mRNA表达水平。结果:与模空组比较,补肾益气活血方高、低剂量组能够增加股骨骨密度(P<0.01),下调血清ALP和TRACP水平(P<0.01),补肾益气活血方高剂量组能够上调Wnt7b/β-catenin信号通路Wnt7b、Axin2和Runx2 mRNA及蛋白表达(P<0.05,P<0.01),补肾益气活血方低剂量组能够上调Wnt7b、Axin2蛋白表达(P<0.01)。结论:补肾益气活血方可以通过上调Wnt7b/β-catenin信号通路以促进成骨分化来防治绝经后骨质疏松症。Objective: Based on Wnt7 b/β-catenin signaling pathway, the effects of Bushen Yiqi Huoxue Formula on femur of postmenopausal osteoporosis rats was studied. Methods: A total of 90 female SD rats were randomly divided into normal group, model blank group, Bushen Yiqi Huoxue Formula high, low dose group and Fosamax group with 18 rats in each group. In addition to the normal group, the rats in the other groups were treated with bilateral ovariectomy to establish the animal model of osteoporosis. The rats in the normal group and the model blank group were given 0.9% sodium chloride solution intragastrically, and the other groups were given the corresponding drugs intragastrically for 12 weeks. The bone mineral density of femur was measured by Dual energy X-ray absorption(DAX), the level of serum ALP, TRACP and the expression of Wnt7 b, Axin2 and Runx2 protein in femur were detected by ELISA, and the expression of Wnt7 b, Axin2 and Runx2 mRNA in femur was detected by qRT-PCR. Results: Compared with the model blank group, Bushen Yiqi Huoxue Formula high, low dose group could increase the bone mineral density of femur(P<0.01) and down-regulate the levels of serum ALP and TRACP(P<0.01). The high dose group of Bushen Yiqi Huoxue Formula could up-regulate the expression of Wnt7 b, Axin2, Runx2 mRNA and protein in the Wnt7 b/β-catenin signaling pathway(P<0.05, P<0.01). The low dose group of Bushen Yiqi Huoxue Formula could up-regulate the expression of Wnt7 b, Axin2 protein(P<0.01). Conclusion: Bushen Yiqi Huoxue Formula can prevent and treat postmenopausal osteoporosis by upregulating Wnt7 b/β-catenin signaling pathway to promote osteogenic differentiation.
