藏药二十五味儿茶凝胶对慢性湿疹大鼠血清中IL-4及IFN-γ表达的影响

摘　　要：目的:观察藏药二十五味儿茶凝胶对大鼠慢性湿疹模型血清中白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)水平的影响。方法:建立大鼠湿疹模型,分为凡士林组、黑豆馏油组及藏药二十五味儿茶凝胶低、中、高(含生药0.14、0.21、0.35g/g)剂量组,常规给药10d后,取大鼠背部造模区域组织做病理观察及炎性细胞计数,采用ELISA法检测各组大鼠血清中IL-4、IFN-γ的表达水平。结果:藏药二十五味儿茶凝胶可不同程度改善大鼠湿疹模型表皮突延长及角化、减轻炎性细胞浸润及细胞间水肿,其中藏药二十五味儿茶凝胶中剂量组使皮肤角质层接近正常;与凡士林组比较,各给药组炎性细胞数显著减少(P<0.05),IL-4表达水平显著降低(P<0.05),IFN-γ表达水平显著升高(P<0.05)。结论:藏药二十五味儿茶凝胶可提高大鼠慢性湿疹模型血清中IFN-γ水平,降低IL-4水平,调节Th1/Th2失衡状态。Objective: To observe the effects of Tibetan medicine Twenty-five Catechins on the serum levels of interleukin-4(IL-4) and interferon-γ(IFN-γ) in chronic eczema rat. Methods: Model rats with eczema were divided into vaseline group, black beans distillation oil group, Tibetan medicine Twenty-five Catechins low, medium and high(including student 0.14, 0.21, 0.35 g/g) dose group, normal dosing after 10 days. The modeling regions on back were taken to do pathological observation and inflammatory cell count, and ELISA method was used to detect serum IL-4, IFN-γ expression levels in each group. Results: Tibetan medicine Twenty-five Catechins could improve epidermal extension and keratinization of rat eczema model, and reduce inflammatory cell infiltration and intercellular edema. Twenty-five Catechins medium group made the stratum corneum close to normal. Compared with vaseline group, inflammatory cells in each administration group were significantly reduced(P<0.05), IL-4 expression level was significantly reduced(P<0.05), and IFN-γ expression level was significantly increased(P<0.05). Conclusion: The Tibetan medicine Twenty-five Catechins can improve the level of IFN-γ level in the serum of chronic eczema rats, reduce the level of IL-4, and adjust the imbalance of Th1/Th2.
