异丹叶大黄素抑制舌鳞癌侵袭转移的研究

摘　　要：目的:研究异丹叶大黄素对舌鳞癌细胞侵袭转移的影响。方法:选取对数期生长的UM1细胞,分为2组,实验组用异丹叶大黄素处理,对照组用PBS处理,采用CCK8实验检测细胞的增殖能力;划痕实验及transwell迁移实验检测2组细胞的迁移能力;transwell侵袭实验测2组细胞的侵袭能力;Western blot检测E-cad、Vimentin、Slug、pERK1/2及ERK1/2的表达情况。结果:异丹叶大黄素对舌鳞癌细胞株UM1的24~48h的半数抑制浓度(IC 50)为:48.2~60.3μmol/L,异丹叶大黄素可以抑制UM1细胞的增殖;异丹叶大黄素抑制舌鳞癌细胞株UM1的迁移及侵袭能力,差异具有统计学意义(P<0.05);UM1细胞经异丹叶大黄素处理后,与对照组相比,Vimentin、Slug、pERK1/2表达下调,E-cad表达升高,ERK1/2表达不变。结论:异丹叶大黄素可抑制舌鳞癌细胞的侵袭转移。Objective:To investigate the effect of Isorhapontigenin(ISO)on migration and invasion of tongue squamous cell carcinoma.Methods:UM1 cells were treated with different concentrations of OA(0,5,10,20,40,60,80 and 100μmol/L)for 24,48h.The cell inhibition rate was evaluated using a modified Cell Counting Kit-8(CCK8)assay.Wound healing assay and transwell assay were performed to analyze the effect of ISO on invasion and migration in UM1 cells.The expression levels of proteins E-cad,Vimentin,Slug,ERK1/2,pERK1/2 were detected by Western blotting.The value P<0.05 was considered statistically significant.Results:ISO inhibits the proliferation of UM1 cells in vitro.The anti-proliferation effect of ISO on UM1 cells was portrayed in a dose-and time-dependent manner.The IC 50 values of coix seed oil were 48.2~60.3μmol/L.ISO inhibited the invasive and migrative ability on UM1 cells(P<0.05).The protein level of Vimentin,Slug,pERK1/2 were decreased with the increasing concentrations of ISO.The expression of E-cad was increased with the increasing concentrations of ISO.ERK1/2 was not obviously changed in UM1 cells afer treatment with ISO.Conclusion:ISO inhibited the migration and invasion of UM1 cells.
