摘 要:【目的】为探讨山羊ASIP、EDN3、PAX3、TYR、TYRP1和DCT 6个黑色素相关基因在不同肤色成年山羊皮肤组织中的表达差异和在B16-F10细胞中的表达规律。【方法】以酉州乌羊和板角山羊皮肤组织、B16-F10增殖和分化细胞为试验材料,应用qRT-PCR检测了以上6个黑色素相关基因的表达并分析了基因间的相关性。【结果】皮肤组织检测结果显示,与板角山羊相比,酉州乌羊皮肤ASIP表达极显著下调(P<0.01),PAX3、TYR、TYRP1和DCT表达显著上调(P<0.05或P<0.01)。体外实验分析结果发现,在黑色素细胞增殖阶段ASIP、EDN3、TYRP1和DCT具有相似的表达模式,而R4X3与7TR表达模式相似,表明它们在色素细胞增殖过程中发挥不同的调节作用。在黑色素细胞分化阶段,ASIP与TYR、TYRP1和DCT具有相似的表达模式,且表达量显著相关,表明他们参与了黑色素细胞的分化及黑色素合成的调控。【结论】ASIP下调与PAX3、TYR、TYRP1、DCT上调与酉州乌羊皮肤黑色素异常沉积表型有密切关系。【Objective】The present paper aimed to study the expression difference of the six melanin-related genes of goat EDN3,PAX3,TYR,TYRPl and DCT in the skin tissues of adult goats with different skin color and the expression pattern in B16-F10 cells.【Meth-od】The skin tissue of Youzhou dark-skin goats and Banjiao goats and the B16-F10 cells of proliferation phase and differentiated phase were used as experimental materials,then the expression levels of six melanin-related genes were detected by qRT-PCR and the correlation of six genes was also analyzed.【Result】The results of skin tissue test showed that compared to Banjiao goats,the expression of ASIP in Youzhou dark-skin goats was significantly down-regulated(P<0.01),but PAX3,TYR,TYRPl and DCT were significantly up-regulated(P<0.05 or P<0.01).The results of vitro experiments showed that in proliferation phase of melanocytes,expression patterns of ASIP,EDN3,TYRPl and DCT were similar,while PAX3 and TYR were similar,indicating that they played different regulatory roles in pigment cell prolif eration phase.In differentiation phase,ASIP had similar expression patterns with TYR,TYRPl and DCT,and the expression levels were sig-nificantly correlated,suggesting that they were involved in the differentiation of melanocytes and the regulation of melanin synthesis.【Conclusion】The down-regulation of ASIP,,the up-regulation of PAX3,TYR,TYRP1 and DCT were closely related to the abnormal depositional phenotype of skin melanin of Youzhou dark-skin goats.
