摘  要:目的评估携带腺病毒介导的肝细胞生长因子基因(Ad-HGF)的脐血间充质干细胞对烧伤后大鼠血浆炎症因子的影响。方法利用密度梯度法分离脐血间充质干细胞,并培养传代。将构建的Ad-HGF转染到脐血间充质干细胞(MOI=100 pfu/cell)。正常大鼠3只,不作处理;将60只大鼠(雌雄对半)建立深II度烧伤模型并随机分为3组:模型对照组(模型组,n=12)、脐血间充质干细胞治疗组(治疗组,n=24)、Ad-HGF修饰的脐血间充质干细胞治疗组(Ad-HGF治疗组,n=24)。将不同药物经尾静脉注射于大鼠体内。于伤后第1、7、14、21天分别检测C反应蛋白(CRP)、血清肿瘤坏死因子(TNF-α)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、集落刺激因子(G-CSF)、白细胞介素-10(IL-10)和白细胞介素-4(IL-4)含量。结果烧伤后第1天,3组大鼠促炎因子(CRP、TNF-α、IL-1和IL-6)和抗炎因子(G-CSF、IL-10及IL-4)水平差异均无统计学意义(P均>0.05);在烧伤后第7、14、21天,治疗组和Ad-HGF治疗组大鼠促炎因子(CRP、TNF-α、IL-1和IL-6)和抗炎因子(G-CSF、IL-10及IL-4)水平均低于模型组,治疗组大鼠促炎因子(CRP、TNF-α、IL-1和IL-6)和抗炎因子(G-CSF、IL-10及IL-4)水平均高于Ad-HGF治疗组(P均<0.01)。结论携带Ad-HGF的脐血间充质干细胞可以调节烧伤后大鼠体内炎症因子的释放,从而减轻炎症反应。Objective To explore the effect of Ad-HGF modified umbilical cord mesenchymal stem cells(UCMSCs)on inflammatory cytokines in burn rats.Methods UCMSCs were isolated by density gradient method and cultured for passage.The constructed AD-HGF was transfected into umbilical cord blood mesenchymal stem cells(MOI=100 pfu/cell).Normal rats were divided into three groups:model control group(model group,n=12),umbilical cord blood mesenchymal stem cell treatment group(treatment group,n=24),and ad-HGF modified umbilical cord blood mesenchymal stem cell treatment group(Ad-HGF treatment group,n=24).Sixty rats(male and female half)were divided into three groups randomly.Different drugs were injected into rats via tail vein.C-reactive protein(CRP),serum tumor necrosis factor(TNF-α),interleukin-1(IL-1),interleukin-6(IL-6),colony-stimulating factor(G-CSF),interleukin-10(IL-10)and interleukin-4(IL-4)were measured on the 1st,7th,14th and 21st day after injury.Results On the first day after burn,there was no significant difference in levels of pro-inflammatory factors(CRP,TNF-α,IL-1 and IL-6)and anti-inflammatory factors(G-CSF,IL-10 and IL-4)among the three groups(P>0.05).On the 7th,14th and 21st day after burn,the levels of pro-inflammatory factors(CRP,TNF-α,IL-1 and IL-6)and anti-inflammatory factors(G-CSF,IL-10 and IL-4)in UCMSCs treatment group and Ad-HGF combined with UCMSCs treatment group were lower than those in saline control group.Meanwhile,the levels of pro-inflammatory factors(CRP,TNF-α,IL-1 and IL-6)and anti-inflammatory factors(G-CSF,IL-10 and IL-4)in UCMSCs treatment group were higher than those in the Ad-HGF combined with UCMSCs treatment group(P<0.01).Conclusion Ad-HGF combined with UCMSCs can inhibit the excessive release of inflammatory cytokines in burn rats.
