摘  要:目的:研究槲皮苷对免疫抑制小鼠免疫功能的调节作用。方法:50只小鼠随机分为对照组、模型组、低剂量组、中剂量组和高剂量组。采用一次性腹腔注射环磷酰胺(CTX)(200 mg/kg)方式建立小鼠免疫抑制模型,对照组腹腔注射等体积生理盐水。低、中、高剂量组分别灌胃20、50、80 mg/kg的槲皮苷,对照组和模型组灌胃等体积生理盐水。14 d后测定胸腺及脾免疫器官指数、腹腔巨噬细胞吞噬百分率及吞噬指数、脾脏组织T淋巴细胞亚群CD4^+、CD8^+亚群及其比例、血浆IgG和IgM、IL-2和IL-4水平并观察脾脏组织病理学状态。以流式细胞术检测抗氧化指标活性氧(ROS),ELISA试剂盒检测丙二醛(MDA)、超氧化物歧化酶(SOD)及过氧化氢酶(CAT)。Western blot检测小鼠脾脏组织核转录因子kappaB(NF-κB)p65和TLR2蛋白表达水平,对槲皮苷的干预作用机制进行探索。结果:与对照组相比,模型组的免疫器官指数、腹腔巨噬细胞吞噬百分率及吞噬指数、CD4^+、CD8^+亚群及其比例、IgG、IgM、IL-2、IL-4、SOD、CAT均显著降低(P<0.05),脾脏病理学损伤明显,而ROS和MDA水平则显著升高(P<0.05);较模型组而言,给予槲皮苷干预后,各组免疫器官指数、腹腔巨噬细胞吞噬百分率及吞噬指数、CD4^+、CD8^+亚群及其比例、IgG、IgM、IL-2、IL-4、SOD、CAT均剂量依赖性地显著升高(P<0.05),脾脏病理学损伤程度呈剂量依赖地减轻,而ROS和MDA水平则剂量依赖性显著降低(P<0.05)。Western blot法检测显示模型组的NF-κB p65和TLR2蛋白均较对照组显著上调(P<0.05);槲皮苷干预各组的NF-κB p65和TLR2蛋白表达水平则剂量依赖性显著下降(P<0.05)。结论:槲皮苷可改善小鼠免疫功能低下状态,通过促进免疫器官功能恢复及提高机体抗氧化水平来提高免疫功能低下小鼠的免疫水平,其作用机制可能与TLR2信号通路有关。Objective:Aim to study the regulation of quercetin on immune function in immunosuppressed mice.Methods:Fifty mice were randomly divided into control group,model group,low dose group,middle dose group and high dose group.The mouse immunosuppressive model was established by intraperitoneal injection of cyclophosphamide(CTX)(200 mg/kg).The control group was intraperitoneally injected with the same volume of normal saline.The low-dose group,the middle-dose group and the high-dose group were intragastrically administrated with 20 mg/kg,50 mg/kg,and 80 mg/kg of quercetin,respectively.The control group and the model group were intragastrically administrated with the same volume of normal saline.After 14 days,the thymus and spleen immune organ index,peritoneal macrophage phagocytosis percentage and phagocytic index,levels of spleen tissue T lymphocyte subsets CD4^+,CD8^+cells and their ratio change,levels of plasma immunoglobulin G,immunoglobulin M,cytokine IL-2 and IL-4 were all measured and the pathological state of spleen was observed.Reactive oxygen species(ROS)were detected by flow cytometry.Malonaldehyde(MDA),superoxide diamutase(SOD)and catalase(CAT)were detected by ELISA kit.Western blot was used to detect the expression levels of nuclear factor kappaB p65 and TLR2 in mouse spleen,and the mechanism of quercetin interfering the inhibited immune function was explored.Results:Compared with the control group,the immune organ index,phagocytosis percentage and index of peritoneal macrophages phagocytosis and phagocytic index,CD4^+,CD8^+cells and their proportion,IgG,IgM,IL-2,IL-4,SOD and CAT were all significantly decreased(P<0.05),spleen pathological damage was significant,while ROS and MDA levels were significantly increased(P<0.05).Compared with the model group,after administration of quercetin,the immune organ index,phagocytosis percentage and index of peritoneal macrophages phagocytosis and phagocytic index,CD4^+,CD8^+cells and their proportion,IgG,IgM,IL-2,IL-4,SOD,CAT were significantly increased in a dose-depen
