摘  要:目的评价在致畸敏感期(妊娠第6~15天)阴道给予苦参凝胶对SD孕鼠的母体毒性、胚胎-胎仔毒性和致畸性。方法将100只孕鼠随机分为4组分别为苦参凝胶高剂量组、中剂量组、低剂量组和对照组(空白凝胶),每组25只。雌性SD大鼠低、中、高剂量组在妊娠第6~15天内分别阴道给予0.5 mL·kg-1·d-1、1 mL·kg-1·d-1、2 mL·kg-1·d-1苦参凝胶,对照组给予1 mL·kg-1·d-1空白凝胶(基质)。试验期间每天观察动物的一般情况和临床毒性表现;记录孕鼠的体质量、摄食量;在妊娠第20天进行剖腹检查,摘取卵巢和子宫,观察黄体数、着床数、胚胎生存和死亡情况、生存胎仔的外观、体质量、骨骼及脏器发育等指标。结果孕鼠及胚胎、胎仔发育的上述各观察指标未见明显异常,与对照组比较无显著性差异。结论在本试验条件下,苦参凝胶未见明显的母体毒性与胚胎-胎仔发育毒性。Objective To evaluate the matrix toxicity,embryo-fetal toxicity,and teratogenicity of the vaginal mucosa irritation of Kushen gel on SD rats in sensitive period to teratogenic agent(day 6~15 of pregnancy).Methods Female rats were administrated with Kushen gel[0.5,1,2 mL/(Kg?d)]at day 6-15 of pregnancy,while the rats in control group were given the stroma of Kushen gel 1 mL/(Kg?d).In the trial period,general conditions and clinical toxicity performance of the animals were observed daily,body weight and food consumption of rats was recorded.SD rats were sacrificed at day 20 after pregnancy.The luteal number,nidation sites,embryo survival and death,the appearance,sex,weight,bone and organ development indicators of survival fetus were observed.Results There were no statistical significance between Kushen gel group and control group in above indexes.Conclusion Kushen gel has no obvious toxicity on embryo and fetus of rats under the conditions in this paper.
