摘 要:目的分析心肌内微小RNA-210(miRNA-210)过表达对急性心肌梗死(AMI)大鼠梗死区域微血管密度和肝细胞生长因子(HGF)表达的影响。方法取40只SD大鼠,随机分为假手术组(缝针穿过心肌,不给予结扎处理)、模型组(结扎冠状动脉前降支建立AMI大鼠模型)、阴性对照组(建立AMI大鼠模型+阴性病毒对照)、实验组(建立AMI大鼠模型+体内转染miRNA-210激动剂),每组各10只。比较各组大鼠术后1个月心肌组织内miRNA-210和HGF表达水平的差异;比较各组大鼠心脏组织病理学改变、心肌组织内微血管密度及超声心动图参数的差异。结果阴性对照组大鼠LVDs较假手术组明显升高,LVFS和LVEF较假手术组、模型组均明显下降(P<0.05);实验组大鼠LVDs较阴性对照组明显下降,LVFS和LVEF较阴性对照组明显升高(P<0.05)。阴性对照组大鼠心肌组织内微血管密度较假手术组明显下降,实验组微血管密度较阴性对照组明显提高(P<0.05)。实时荧光定量聚合酶链反应结果显示,相比假手术组、模型组和阴性对照组,实验组大鼠HGF与miRNA-210表达水平显著升高(P<0.05);相比假手术组和阴性对照组,模型组大鼠miRNA-210表达水平显著升高(P<0.05)。免疫印迹法检测结果显示,实验组大鼠HGF蛋白的表达水平明显高于其余各组,差异有显著性(P<0.05)。结论心肌组织内miRNA-210过高表达可促使AMI大鼠梗死区域微血管密度和HGF表达升高,因此可起到诱导血管新生和改善心脏收缩功能的作用。Objective To analyze the effect of microRNA-210(miRNA-210)overexpression on microvessel density and hepatocyte growth factor(HGF)expression in infarcted area of acute myocardial infarction(AMI)rats.Methods Forty SD rats were randomly divided into sham operation group(suture needle penetrating myocardium without ligation),model group(establishment of AMI rat model),negative control group(establishment of AMI rat model+negative virus control),experimental group(establishment of AMI rat model+in vivo transfection of miRNA-210 agonist),10 rats in each group.The expression levels of miRNA-210 and HGF in myocardial tissues of rats in each group at 1 month after operation were compared.The pathological changes,microvessel density and echocardiographic parameters in myocardial tissues of rats in each group were compared.Results LVDs in the negative control group were significantly higher than those in the sham-operated group,LVFS and LVEF were significantly lower than those in the sham-operated group and model group(P<0.05).LVDs in the experimental group were significantly lower than those in the negative control group,LVFS and LVEF were significantly higher than those in the negative control group(P<0.05).The microvessel density in the myocardium of the negative control group was significantly lower than that of the sham-operated group,and the microvessel density of the experimental group was significantly higher than that of the negative control group(P<0.05).Real-time fluorescence quantitative polymerase chain reaction showed that compared with sham-operated group,model group and negative control group,the expression levels of HGF and miRNA-210 in experimental group were significantly higher(P<0.05);compared with sham-operated group and negative control group,the expression levels of miRNA-210 in model group were significantly increased(P<0.05).Western blot analysis showed that the expression level of HGF protein in the experimental group was the most significant in each group.Conclusion Overexpression of miRNA-210 in
