摘 要:目的研究胰高血糖素样肽-1(GLP-1)对糖尿病(DM)模型大鼠肾脏的抗炎及抗氧化作用及对p38MAPK通路的影响。方法选择成年雄性SD大鼠并随机分为对照组、DM组、GLP-1组,后两组采用链脲佐菌素腹腔注射的方式建立DM模型,GLP-1组给予GLP-1腹腔注射干预、连续8周。比较三组间肾功能指标、炎症指标[核因子κB(NF-κB)、白细胞介素(IL)-1β、IL-17、肿瘤坏死因子α(TNF-α)]、氧化应激指标[一氧化氮(NO)、诱导型一氧化氮合成酶(iNOS)、活性氧(ROS)、超氧化物歧化酶(SOD)]、p38MAPK通路基因的差异。结果DM组大鼠的血肌酐、血尿素、24 h尿蛋白水平及肾脏中NF-κB、IL-1β、IL-17、TNF-α、ROS、NO、iNOS的含量、p38MAPK、Caspase-3的表达水平明显高于对照组,肾脏中SOD、GSH-Px的含量及PPARγ的表达水平明显低于对照组;GLP-1组大鼠的血肌酐、血尿素、24 h尿蛋白水平及肾脏中NF-κB、IL-1β、IL-17、TNF-α、ROS、NO、iNOS的含量、p38MAPK、Caspase-3的表达水平明显低于DM组,肾脏中SOD、GSH-Px的含量及PPARγ的表达水平明显高于DM组。结论GLP-1对糖尿病模型大鼠肾脏具有抗炎及抗氧化作用,且该作用与p38MAPK通路的抑制有关。Objective To study the anti-inflammatory and antioxidant effects of glucagon-like peptide-1(GLP-1)on the kidney of diabetic rats and its effect on p38MAPK pathway.Methods Adult male SD rats were randomly divided into control group,DM group and GLP-1 group.DM model was established by intraperitoneal injection of streptozotocin in the latter two groups.GLP-1 group was intervened by intraperitoneal injection of GLP-1 for 8 weeks.The differences of renal function index,inflammatory index,oxidative stress index and p38MAPK pathway gene among the three groups were compared.Results The levels of serum creatinine,blood urea,24 hour urinary protein and the contents of NF-κB,IL-1β,IL-17,TNF-α,ROS,NO,iNOS,the expression levels of p38MAPK and Caspase-3 in renal tissues of DM group were significantly higher than those in control group,while the contents of SOD,GSH-Px and the expression levels of PPAR-γin renal tissues were significantly lower than those in control group.The levels of serum creatinine,blood urea,24-hour urinary protein and the contents of NF-κB,IL-1β,IL-17,TNF-α,ROS,NO,iNOS,the expression levels of p38MAPK and Caspase-3 in renal tissues of GLP-1 group were significantly lower than DM group,while the contents of SOD,GSH-Px and the expression levels of PPAR-γin renal tissues were significantly higher than those in DM group.Conclusion GLP-1 has anti-inflammatory and antioxidant effects in the kidneys of diabetic rats,and this effect is related to the inhibition of p38MAPK pathway.
