摘  要:目的探讨莪术醇对子宫腺肌症异位子宫内膜间质细胞(EESCs)细胞增殖、凋亡及迁移的影响,为临床治疗子宫腺肌症提供理论依据。方法对EESCs进行分离、纯化培养及细胞鉴定。将传至第2—3代的EESCs细胞随机分为5组:Control组(加入质量浓度为10 mg·L-1的无水乙醇作用于EESCs)和莪术醇1、2、3、4组(分别加入质量浓度为12.50、25.00、50.00、100.00 mg·L-1的莪术醇作用于EESCs)。采用CCK-8法检测EESCs的细胞存活率,细胞划痕实验检测EESCs的细胞迁移率,流式细胞术联合FITC-Annexin V/PI双染色法检测EESCs的细胞凋亡率。结果分别用鼠抗人vimentin、cytokine抗体鉴定EESCs显示细胞纯度在95%以上,成功分离培养出原代EESCs细胞。与Control组比较,莪术醇1、2、3、4组24、48 h时EESCs细胞增殖率、24 h EESCs细胞迁移率均降低,EESCs细胞凋亡率均升高,差异有统计学意义(P<0.05)。莪术醇1、2、3、4组24、48及72 h时EESCs细胞增殖率均低于96 h,莪术醇3、4组EESCs细胞凋亡率均较莪术醇1、2组升高,莪术醇4组EESCs细胞凋亡率较莪术醇3组升高,差异有统计学意义(P<0.05)。结论莪术醇在体外能显著抑制子宫腺肌症EESCs细胞增殖和迁移,诱导细胞凋亡,具有潜在的治疗子宫腺肌症作用。Objective To explore the effects of curcumol on proliferation,migration and apoptosis of ectopic endometrial stromal cells(EESCs),and to provide a theoretical basis for the treatment of adenomyosis.Methods EESCs were isolated,purified,cultured and identified.The cells at passage 2-3 were randomly treated with 10 mg·L-1 anhydrous ethanol(control group),12.50 mg·L-1 curcumol(curcumol group 1),25.00 mg·L-1 curcumol(curcumol group 2),50.00 mg·L-1 curcumol(curcumol group 3),or 100.00 mg·L-1 curcumol(curcumol group 4),The proliferation,migration and apoptosis of EESCs were measured using CCK-8 assay,scratch analysis and FITC-Annexin V/PI flow cytometry,respectively.Results Primary EESCs were isolated and cultured successfully,and the identification for anti-vimentin and anti-cytokine antibodies showed that the cell purity was above 95%.Compared with control group,the proliferation rates were reduced after treatment for 24 and 48 hours,the migration rates were decreased after treatment for 24 hours,and the apoptosis rates were elevated in all the four curcumol groups(P<0.05).Furthermore,the proliferation rates after treatment for 24,48 and 72 hours were higher than those after treatment for 96 hours in curcumol groups(P<0.05).Compared with curcumol group 1 and group 2,the apoptosis rates increased in curcumol group 3 and group 4 in a dose-dependent manner(P<0.05).Conclusion Curcumol treatment has a potential therapeutic effect on adenomyosis by inhibiting proliferation and migration and inducing apoptosis in EESCs.
