摘　　要：目的:研究丹莪妇康煎膏含药血清对子宫腺肌病间质细胞Y14的细胞生物学功能的影响。方法:制备丹莪妇康煎膏含药血清,作用于间质细胞Y14,CCK-8、Transwell及流式细胞术检测细胞增殖、迁移、侵袭及凋亡情况;同位素相对标记与绝对定量技术检测差异表达蛋白情况。结果:与对照组相比,丹莪妇康煎膏含药血清高剂量组作用96h,抑制细胞增殖差异显著(P<0.05);中、高剂量组细胞迁移数及侵袭数均显著下降(P<0.05);同位素相对标记与绝对定量技术检测并筛选到差异表达蛋白质共188个,其中表达上调的蛋白质81个,表达下调的蛋白质107个。结论:丹莪妇康煎膏能抑制Y14细胞的增殖、迁移及侵袭;同位素相对标记与绝对定量技术筛选出的差异表达蛋白质为进一步研究其作用机制提供了基础。Objective:To investigate the molecular mechanism that DanE Fukang Decoction plays role on ectopic endometrial stromal cells Y14 isolated from adenomyosis.Methods:Serum containing DanE Fukang Decoction obtained from female Sprague Dawley rats intragastrically administered with DanE Fukang Decoction.Proliferation,apoptosis,migration and invasion of Y14 were detected after treated with serum containing DanE Fukang Decoction,via CCK-8,flow cytometry and transwell.iTRAQ technology was used to detect differentially expressed proteins.Results:Compared with the control group,the high-dose group was significantly inhibit cell proliferation at 96h(P<0.05),the number of cell migration and invasion in the middle and high dose groups decreased significantly(P<0.05).iTRAQ technique detected and screened differentially expressed proteins 188,which up-regulated expression proteins 81,and down-regulated expression proteins 107.Conclusion:Serum containing DanE Fukang Decoction could inhibit the Y14 cells proliferation,migration and invasion.The differentially expressed proteins detected via iTRAQ provide a basis for further study of its mechanism.
