摘 要:目的分析T淋巴细胞免疫球蛋白黏蛋白域分子-3(TIM-3)抑制对支气管哮喘患者免疫功能的影响。方法选取2013年8月—2015年8月收治的64例支气管哮喘患者(急性发作期32例,缓解期32例)作为观察组,另选取32例健康受试者作为对照组。比较2组以及急性发作期和缓解期支气管哮喘患者血清IL-10、IL-17水平,比较2组TIM-3抑制前后外周血中Th17和Treg细胞比例、外周血中TIM-3表达情况以及外周血CD4+T细胞中TIM-3的表达水平。结果观察组血清IL-10和IL-17水平以及外周血CD4+T细胞中TIM-3的表达均显著高于对照组(P<0.01)。急性发作期患者的血清IL-10和IL-17水平均明显高于缓解期患者(P<0.01)。TIM-3抑制前,观察组外周血Th17细胞比例以及Th17/Treg均高于对照组,Treg细胞比例低于对照组(P<0.05)。TIM-3抑制后,观察组外周血Th17细胞比例以及Th17/Treg均较抑制前降低,Treg细胞比例较抑制前升高,且Th17细胞比例以及Th17/Treg明显高于对照组(P<0.05)。结论TIM-3抑制能有效调节支气管哮喘患者Th17和Treg细胞的平衡,减轻患者的气道炎症,其疗效显著。Objective To analyze effect of TIM-3 inhibition on immune function in patients with bronchial asthma.Methods A total of 64 patients with bronchial asthma(32 patients in acute stage and 32 patients in remission stage)admitted during August 2013 and August 2015 were selected as observation group,and another 32 healthy subjects were selected as control group.Serum interleukin-10(IL-10)and interleukin-17(IL-17)levels were compared between two groups,and the patients with acute stage and remission bronchial asthma.Ratio of Th17 to Treg cells in peripheral blood and TIM-3 expression in peripheral blood before and after TIM-3 inhibition and TIM-3 expressions in CD4+T cells in peripheral blood were compared in two groups.Results Values of serum IL-10 and IL-17 levels and TIM-3 expression in CD4+T cells in peripheral blood were significantly higher in observation group than those in control group(P<0.01).Serum IL-10 and IL-17 levels in patients with acute stage were significantly higher than those in patients with remission stage(P<0.01).Before TIM-3 inhibition,values of ratio of Th17 cells and Th17/Treg in peripheral blood in observation group were significantly higher while ratio of Treg cells was significantly lower than those in control group(P<0.05).After TIM-3 inhibition in observation group,values of ratio of Th17 cells and Th17/Treg in peripheral blood were significantly lower,while ratio of Treg cells was significantly higher than those before inhibition,and improved from those before the inhibition,and values of ratio of Th17 cells and Th17/Treg were significantly higher than those in control group(P<0.05).Conclusion TIM-3 inhibition may effectively regulate the balance of Th17 and Treg cells and reduce airway inflammation with significant curative effect in treatment of patients with bronchial asthma.
