摘　　要：为鉴定一种以鸡心包积液、肝炎为主要临床症状疾病的病原,采集3份发病鸡肝脏组织,接种9日龄SPF鸡胚分离病毒,并进行PCR鉴定及序列分析。结果表明,鸡胚尿囊液中检测出大小约1 032 bp的目的片段,序列比对分析显示,分离的病毒与禽腺病毒C种4型同源性最高,达98.30%~98.80%。理化特性分析表明,病毒无血凝性,对BUDR敏感,对氯仿、乙醚不敏感。中和试验证实,4型禽腺病毒阳性血清能够有效中和病毒的感染,分离病毒攻毒SPF雏鸡后能够完全复制出与临床自然感染鸡相同的症状,病理组织学结果显示肝脏组织切片出现大量嗜碱性核内包涵体,心肌纤维间隙炎性细胞浸润。说明分离得到的病毒为禽腺病毒Ⅰ群4型。In order to identify a kind of disease characterized by pericardial effusion and hepatitis,hepatic samples were collected and the virus was isolated by SPF chicken embryos.PCR and sequence analysis were used to identify the virus.The result showed that a 1 032 bp gene was detected in SPF chicken embryos.Sequence analysis showed that isolated stains were closest to fowl adenovirus 4(98.30%-98.80%).The clinical signs and lesions were reproduced in SPF chickens via experimental infection with the isolated virus.The results of physical and chemical property test showed that the isolated virus was not hemagglutinating,insensitive to chloroform and diethyl ether,and resistant to acid.Neutralization experiments confirmed that the positive sera of FAdV-4 were able to effectively neutralize viral infections.The SPF chick challenge test confirmed that the isolated virus can completely replicate the same symptoms and lesions as the naturally occurring chickens.The result of histopathology showed the obvious basophilic nuclear inclusions in the liver and inflammatory cell infiltration between myocardial fibers.The above results indicate that the virus isolated from the diseased chickens is fowl adenovirus serotype 4.
