摘  要:为探讨玉屏风多糖脂质体(YPF-PL)的免疫调节活性,研究其对呼吸道和肠道黏膜免疫功能的影响,以180羽13日龄SPF麻黄鸡作为试验动物,随机分为6组,每组30羽,即玉屏风多糖脂质体高、中、低剂量组(400、200、100 mg/kg/羽),玉屏风多糖对照组(200 mg/kg/羽),免疫对照组和空白对照组,混合饲料给药,连续给药7 d,给药第4天免疫接种禽流感病毒H5N1灭活疫苗;28日龄以同样方式二次给药和二次免疫,42日龄每组随机选10羽处死。试验期间每周采血获得血清,采用HI法检测每周各组雏鸡抗体水平变化,收集肺脏和肠道分泌液,竞争ELISA检测各组肺脏、肠道黏膜免疫主要效应蛋白SIgA含量变化;取空肠和肺组织石蜡包埋、HE染色,光学显微镜下观察肺脏病理组织学变化、空肠杯状细胞、肠绒毛固有层淋巴细胞和黏膜上皮内淋巴细胞变化,测量隐窝深度和绒毛长度,统计V/C值。结果发现,高剂量玉屏风多糖脂质体可增强禽流感疫苗免疫效果,能较长时间维持高水平抗体效价;显著促进肺脏、肠道SIgA分泌(P<0.01);能促进肺脏支气管相关淋巴组织增生,引起肺淋巴细胞归巢明显;空肠杯状细胞、固有层淋巴细胞和黏膜上皮内淋巴细胞数量显著上升(P<0.01),绒毛高度和V/C比值显著升高(P<0.01)。结果提示玉屏风多糖脂质体能够促进雏鸡禽流感疫苗免疫效果,增强机体呼吸道黏膜免疫功能和肠黏膜免疫功能,作用效果优于玉屏风多糖。By studying the effects of Yupingfeng polysaccharide liposomes on the immune function of respiratory and intestinal mucosa,to investigate the immunological activity of Yupingfeng polysaccharide liposomes and evaluate the advantages of Yupingfeng polysaccharide liposomes,180 13-day-old SPF chickens were randomly divided into 6 groups;then for 7 days,Yupingfeng polysaccharide group and high,medium and low dose Yupingfeng polysaccharide liposome group were administered at a dose of 200 mg/kg,400 mg/kg,200 mg/kg and 100 mg/kg respectively,other two control groups were given an equal volume of normal saline;The chickens were injected with avian influenza vaccine under the skin in the fourth day of administration except the blank control group.Second immunization was in the 28-day-old chickens.During the test,blood was collected every week to obtain serum,and the levels of antibody in each group of chicks were measured by HI method.At the age of 42 days,10 chickens were randomly selected and killed,and the lung and intestinal secretions were collected.The contents of SIgA in the lung and intestinal mucosa of each group were detected by competitive ELISA.The jejunum and lung tissue were used to make sections for HE staining,then the histological changes of the lungs,jejunal goblet cells,intestinal villus lamina propria lymphocytes and intraepithelial lymphocytes were observed;Next,the crypt depth and the villus length were measured,and the V/C ratio was counted.High-dose Yupingfeng polysaccharide liposomes can enhance the immune effect of avian influenza vaccine and maintain high levels of antibody titers for a long time,significantly promote lung and intestinal SIgA secretions(P<0.01),promote the proliferations of lung and bronchial-associated lymphoid tissue,cause obvious homing of lung lymphocytes;besides,the number of goblet cells,lamina propria lymphocytes and intraepithelial lymphocytes increased(P<0.01),and the villus height and V/C ratio increased significantly(P<0.01).Yupingfeng polysaccharide liposomes can pro
