摘 要:探寻密花石豆兰中石豆菲醌的质量控制方法。方法采用有机溶剂超声提取的方法提取菲醌,建立硅胶薄层色谱的鉴别方法和高效液相色谱定量分析方法,以Agilent Eclipse XDB-C 18(4.6 mm×250 mm,5μm)为色谱柱,流动相为乙腈-水溶液(60∶40),流速为1.0 mL/min,检测波长为266 nm;进样体积:10μL。结果:通过薄层色谱初步鉴别石豆菲醌后进行HPLC定量分析,其测定浓度在50~500μg·mL^-1范围内与峰面积呈良好的线性关系(r=0.9892),平均回收率为99.41%,RSD为1.6%,n=6。结论:该法操作简便、准确、灵敏,可用于密花石豆兰的石豆菲醌质量控制。Objective To explore the quality control method of monoterpenylbenzoquinone in Bulbophyllum odoratissimum.Method:Phenanthrenequinone was extracted by ultrasonic extraction of organic solvents.The identification method of silica gel TLC and the quantitative analysis method of high performance liquid chromatography were established.The separation was carried out on Agilent Eclipse XDB-C 18(4.6 mm×250 mm,5μm)column with acetonitrile-water(60∶40)as mobile phase at the flow rate of 1.0 mL·min^-1.The UV detection wavelength was set at 266 nm and the injection volume was 10μL.Result:Quantitative analysis of Phenanthrenequinone by HPLC was carried out after identification of Phenanthrenequinone by TLC.The linear range of chrysin was 5~500μg·mL^-1(r=0.9892)with an average recovery of 99.41%(RSD1.6%,n=6).Conclusion:The method is accurate,reliable and reproducible,and it is can be used for the content determina-tion of Bulbophyllum odoratissimum.
