摘 要:目的:观察加味涤痰汤对血管性轻度认知障碍(vascular mild cognitive impairment,VaMCI)模型大鼠脑皮质白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、一氧化氮(nitric oxide,NO)水平及一氧化氮合酶(nitric oxide synthase,NOS)的影响。方法:将48只大鼠按照随机数字表法分为假手术组、模型组、加味涤痰汤组和尼莫地平组,每组各12只。采用在大鼠右侧颈总动脉结扎结合半高脂饲料喂食8周的方法,构建血管性轻度认知障碍模型,测定模型大鼠脑皮质IL-6、TNF-α水平及NO含量、NOS活性的影响。结果:与假手术组比较,模型组大鼠第8周参考记忆错误和工作记忆错误次数增加,差异具有统计学意义(P<0.05)。与模型组比较,加味涤痰汤组大鼠第8周时参考记忆错误和工作记忆错误次数均显著减少,尼莫地平组大鼠参考记忆错误次数明显减少,差异具有统计学意义(P<0.05)。与假手术组比较,模型组大鼠大脑皮质IL-6、TNF-α、NO含量、NOS活力升高(P<0.05);与模型组比较,加味涤痰汤组及尼莫地平组大鼠脑皮质IL-6、TNF-α、NO含量、NOS活力下降(P<0.01);与尼莫地平组比较,加味涤痰汤组大鼠脑皮质IL-6、TNF-α、NO含量、NOS活力差异无统计学意义(P>0.05)。结论:加味涤痰汤能够改善模型大鼠认知功能,通过降低模型大鼠脑皮质IL-6、TNF-α、NO的含量及NOS活性起到神经保护作用。Objective:To observe the influence of Supplemented Phlegm-Flushing Decoction on the levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),nitric oxide(NO)and nitric oxide synthase(NOS)activit in vascular mild cognitive impairment(VaMCI)model rats.Methods:A model rat of VaMCI was established by ligating the right common carotid artery and feeding with semi-high fat diet for 8 weeks.The levels of IL-6,TNF-α,NO content and NOS activity in cerebral cortex were measured.Results:Compared with the sham-operated group,the numbers of reference and working memory errors increased in the model group at the 8th week,and all the differences were statistically significant(P<0.05).Compared with the model group,the numbers of reference memory enors and working memory errors decreased significantly in the group of Supplemented Phlegm-Flushing Decoction at the 8th week,and the number of reference memory errors decreased significantly in the Nimodipine group,and all the differences were statistically significant(P<0.05).Compared with the sham-operated group,the contents of IL-6,TNF-α,NO and NOS activity in cerebral cortex of rats in the model group increased(P<0.05).Compared with the model group,the contents of IL-6,TNF-α,NO and NOS activity in cortex of rats in the group of Supplemented Phlegm-Flushing Decoction and the Nimodipine group decreased(P<O.01).Compared with the Nimodipine group,there was no significant difference in IL-6,TNF-α,NO content and NOS activity in cortex of rats between the group of Supplemented Phlegm-Flushing Decoction and the Nimodipine group(P>0.05).Conclusion:Supplemented Phlegm-Flushing Decoction can improve the cognitive function of model rats,and play a neuroprotective role by reducing the contents of IL-6,TNF-α,NO and NOS activity in cerebral cortex of model rats.
