摘 要:为了调查研究河南省某规模化养猪场粪肠球菌中酰胺醇类-噁唑烷酮类交叉耐药基因optrA、cfr和poxtA的扩散情况,对52份采样菌株进行了分离、纯化和16S rRNA基因序列测序,并进行了药物敏感性测定、接合转移及MLST分型试验。通过16S rRNA基因序列测序比对,鉴定52份采样菌株为粪肠球菌,序列同源性达99.9%。药物敏感性实验结果表明,52株粪肠球菌分离株对氟苯尼考的耐药率达76.9%,对利奈唑胺的耐药率为9.6%(5/52)。PCR扩增结果显示optrA阳性检出率为75.00%(39/52),poxtA阳性检出率为0.19%(1/52),cfr阳性检出率为0%。通过平板接合试验获得了4株optrA阳性接合子,发现其耐药表型发生了转移。多位点序列分型(MLST)试验结果表明:在接合成功的4株供体野生菌株中,3株(水源/病猪源/苍蝇源)分型为ST692,1株仔猪源野生株分型为ST632。In order to investigate the spread of cross-resistance genes optrA,cfr and poxtA in a large-scale swine farm in Henan Province.Methods:The 52 isolated strains were isolated and purified,and the 16S rRNA gene sequencing,drug sensitivity test,joint transfer test and MLST typing test were performed.The 52 strains were identified as Enterococcus faecalis with sequence homology of 99.9% by sequencing of the 16S rRNA gene.Drug sensitivity experiments showed that the resistance rate of 52 strains of Enterococcus faecalis to florfenicol was 76.9%,and the resistance rate to linezolid was 9.6%(5/52).A total of 39 optrA positive strains were detected by PCR.The positive rate of optrA was 75%(39/52),and only one strain of poxtA positive strain was detected.The positive detection rate of Enterococcus faecalis was 0.19%(1/52).The cfr positive strain was not detected,and the positive detection rate was 0%.Four optrA-positive zygote were obtained by plate-joining test,and the transfer of drug-resistant phenotype was found.Multi-site sequence typing(MLST)showed that among the four donor wild strains,three strains(water source/sick pig/fly)were typed ST692,and one piglet wild type was ST632.
