摘 要:目的观察不同针刺施术时长对脑出血模型大鼠脑组织炎症因子及应激蛋白的影响。方法将40只SD大鼠随机分为假手术组、模型组、针刺5 s组、针刺60 s组,每组10只。模型组、针刺5 s组、针刺60 s组复制脑出血模型,其中针刺5 s、60 s组取大鼠"水沟""内关"穴施雀啄、捻转手法5 s或60 s,每日1次。假手术、模型组不予针刺。观察干预5 d后脑血肿区域细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)免疫组化含量及热休克蛋白(HSP-70)的蛋白表达水平。结果 1)与假手术组相比,模型组、针刺5 s、60 s组的ICAM-1、TNF-α、IL-1β、IL-6、HSP-70表达升高(P <0.05);2)针刺60 s组的ICAM-1、HSP-70含量较模型组、针刺5 s组改善更明显(P <0.05);3)针刺5 s、60 s组的TNF-α、IL-1β、IL-6含量较模型组降低,且针刺60 s组的TNF-α、IL-6降低更显著(P <0.05)。结论针刺对改善大鼠炎症反应、保护脑组织有效且针刺60 s组的各项指标改善明显,较长针刺施术时长对抑制大鼠炎症因子表达、减轻脑损伤有正向作用。Objective:To explore the effects of different acupuncture techniques on inflammatory factors and heat shock protein 70(HSP-70)in brain tissue of rats with cerebral hemorrhage.Methods:40 SD rats were randomly divided into sham operation group,the model group,acupuncture 5 s group and acupuncture 60 s group,10 rats in each.In the model group and acupuncture groups,the cerebral hemorrhage model was replicated.In the acupuncture groups,rats in the"shuigou"and"neiguan"acupoints were applied with pecking and twisting manipulation for 5 s or 60 s,once a day.No acupuncture was performed in the sham and model groups.The immunohistochemical levels of ICAM-1,TNF-α,Il-1β,Il-6 and HSP-70 were observed in the hematoma area after intervention for 5 days.Results:Compared with the sham group,the expressions of ICAM-1,TNF-α,Il-1β,Il-6 and HSP-70 in the model group,acupuncture group,5 s group and 60 s group were increased(P<0.05).The levels of ICAM-1 and HSP-70 in 60s group were significantly improved compared with the model group and 5s group(P<0.05).Compared with the model group,the contents of TNF-α,Il-1βand Il-6 in 5 s and 60 s acupuncture groups were decreased,and the decreases of TNF-α and Il-6 in 60s acupuncture group were more significant(P<0.05).Conclusion:Acupuncture is effective in improving inflammation and protecting brain tissue in rats,and the indexes in the 60s group are improved obviously.The longer time of acupuncture application has a positive effect on inhibiting the expression of inflammatory factors and alleviating brain injury in rats.
